Purpose:In this study, Rapid resolution liquid chromatography-electrospray ionization massspectrometry (RRLC-Q-TOF) analyze the chemical composition of the flower of Puerarialobata (Willd.) Ohwi. The flower of Pueraria lobata (Willd.) Ohwi which is short for Ge Huain Chinese, plays a important role about the protective effect of relieving alcoholism andprotecting liver in traditional Chinese medicine. The study also analyses the main chemicalcompound of ginseng Panax ginseng C.A Mey.The active chemical compounds to be analysisin vitro and in vivo.Using principal component analysis (PCA) to differentiate the statisticaldata.By studying the Chinese medicine theory, we hope the mechanism of safety and effectiveproducts can be study later.Method:1. Sample-making:Ge Hua,ginseng1:1mix weighing, adding eight times the amount of water,after1h boiling,filtration;adding a second five times the amount of boiling water, boiling30min after thefilter to extract decoction, twice the combined filtrate is Gehua ginseng decoction;Gehuasingle decoction and ginseng single decoction prepared as above.Extraction of ginsenosides in2010according to the Pharmacopoeia of China, according to the water extraction and alcoholprecipitation ginseng extract polysaccharides.The decoction prepared to stand by10000r min-1centrifuge10min, and get the supernatant,0.22μm filter membrane,4℃preservation.2. LC-MS analysis:Testing Ge Hua and ginseng samples by rapid resolution liquid chromatography quadrupoletime-of-flight mass spectrometry (RRLC/Q-TOF-MS), we can obtain the information ofchromatography.3. Alcohol dehydrogenase (ADH) enzyme activity experiment:Applications Valle-Hooch (Valle&Hoch) activity was measured in vitro effects of drugs onalcohol dehydrogenase,and long-term tests on mice Gehua ginseng drink alcoholic liveralcohol dehydrogenase (ADH) changes.4. Data analysis: Principal component analysis (PCA) to the experimental data analysis, study differenceschromatographic behavior between Ge Hua with ginseng and ginseng single decoction.Result:1. In the RRLC/Q-TOF-MS analysis Gehua water extraction, the main component ofisoflavones and their glycosides, saponins and phenols concurrent presence in Gehua.Ge wasfirst discovered in the same plant kudzu Pueraria alopecuroides Craib bristlecone uniquecompounds alopecuroides A and3’-methoxy daidzein-7-o-methylether.2. In the study of the compatibility of Ge Hua Ginseng, PCA score plot shows that Ge Huawith ginseng and ginseng single decoction decoction mixed completely separated, the resultsare displayed in the chemical composition and content of the single and mixed decoctiondecoction significantly different.3. The alcohol dehydrogenase enzyme activity experiment, ginsenoside and extraction ofGehua can promote reaction of ADH. By contrast, ginseng polysaccharides can inhibitionreaction of ADH.Result shows that extraction of Gehua with ginseng has a protective effect oflong-term consumption of alcoholic liver.Conclusion:1. The main chemical components of Ge Hua are kakkalide, irisolidone, tectoridi, tectorigenin,6"-O-xylosyl-tectoridin and genistin and so forth. Due to using of water extract method,somecompounds are not soluble in water.In this study, it is the first time to discovered theAlopecuroides A in the kudzu.Most compounds of isoflavones in MS,a huge distinctionbetween isoflavones and flavonoids, such as major fragmentation pathway of flavonoids isRDA fracture.No matter isoflavones or flavonoids in fragmentation behavior, they are botheasy to lose C ring and bond C-O.Easy to lose methoxy methylated.The identification of thesefeatures of Gehua provide accurate and rapid method for analysis.2. Boiling with Ge Hua and ginseng, ginseng can promote Gehua dissolution of isoflavones,while ginseng ginsenosides has not diminished, but increased.The results may be due to thestructure of the molecules leads to increased dissolution.The results of alcohol dehydrogenaseactivity experiments shows that the activity of the order of Decoction>mixture≈Gehua singledecoction>ginseng single decoction, and compatibility with both the role of the material must be consistent with the results-based research.Gehua compatibility with ginseng in vivo and invivo both play the important role in relieving alcoholism and protecting liver.
