Protective Effect Of Pueraria Lobata Extract On Mice With Alcoholism

Alcoholism refers to a mental or pHysical health problem caused by excessive drinking.It mainly causes serious damage to the liver and brain nervous system.The clinical manifestation includes alcoholic liver disease and behavioral awareness disorder,usually accompanied by alcohol addiction and learning and memory questions.The pathogenesis of alcoholism is complicated,and there is a lack of clinically available drugs for the prevention and treatment of alcoholism so far.The main active ingredient of Pueraria lobata is isoflavones,and its pHarmacological effects are very extensive.The ancient medical book "Compendium of Materia Medica" records that puerarin has the effect of sobering up and liver protection,but currently there are few clinical studies and applications,and the specific effects and mechanisms of action need to be explored.In this experiment,the protective effects of Pueraria lobata extract on acute alcoholism and chronic alcoholism were studied,and the mechanism of action was explored.The specific work was as follows:(1)In this experiment,a mouse model of acute alcoholism was established by intragastric administration of a certain concentration of alcohol,and then the pretreatment of Pueraria lobata extract was used to explore its effect and mechanism.It was found that Pueraria lobata extract could prolong the time of intoxication and shorten the time of sobering up;Then it could reduce the alcohol concentration in the blood,and also improve the activity of ADH and ALDH in the liver of mice,besides effectively reduce the level of ?-EP and DOR in the brain tissue of mice;in addition,pathological detection also indicated that it has a certain protective effect on brain tissue and liver tissue damage.(2)In order to further explore the effect of puerarin extract on liver damage caused by chronic alcoholism and its mechanism of action,we established a model of chronic alcoholic liver injury and tested the body weight,liver index,serum lipid levels and antioxidant levels of mice.The results showed that Pueraria lobata extract improved the body weight,liver index and liver function indexes AST,ALT and ALP abnormalities.It also down-regulated the alcohol-induced blood lipid indexes TC and TG,and effectively improved the antioxidant index.The levels of GSH,GST and T-AOC significantly enhanced ADH activity in the liver.(3)Finally,we studied the protective effect of Pueraria lobata extract on chronic alcoholism brain damage.The results showed that Pueraria lobata extract effectively reduced the expression of caspase-3 in cerebral cortex;behavioral tests indicated that Pueraria lobata extract could improve learning and memory dysfunction in mice created by chronic alcohol;in addition,Pueraria lobata extract promoted the expression of TH in the midbrain and its changes were related to learning and memory function.The above results show that alcohol can cause serious damage to the liver and brain tissue of the body.Pueraria lobata extract has protective effects on acute alcoholism and chronic alcoholism,and effectively improves alcohol damage.The specific mechanism of action may be related to the activity of alcohol metabolism enzymes increasing,the levels of antioxidants raising and the delta opioid receptor system in the brain.
