Objective: To detect PI3KC3 and Beclin1 expression of liver cancer tissue and liver tissue in rats, explore the role of PI3KC3 and Beclin1 in the occurrence and development of primary hepatic carcinoma and their correlation.Methods: Sixty male Sprague-Dawley rats were randomly divided into the Hepatocellular Carcinoma(HCC) group(n=50) and control group(n=10). The rats in HCC group were treated with 4 mg/ml DEN(25 mg/kg) by intraperitoneal injection three times every week, and the rats in control group were treated with same amount of saline by intraperitoneal injection. The liver space-occupying nodule was confirmed by B-mode ultrasonographyfor in 16 weeks. Put to death all the rats in 18 weeks, the liver cancer tissue samples and the normal liver tissue samples were collected for HE staining. To judge the success of liver cancer model in rats,the rat liver nodules of the HCC group and the rat liver tissues of control group were stained by HE. The rats liver cancer specimens were divided into 4 classifications by Edmondson, because of the different degree of differentiation. The PI3KC3 and Beclin1 expressions were detected in HCC group and control group by RT-PCR and immunohistochemical method. Using single factor analysis of variance, significant difference among groups in terms of analysis and Pearson correlation analysis for correlation test.Results:1. In 18 weeks we put to death all the rats,including liver cancer group 34 and the control group 8. 30 liver cancer tissue samples of the liver cancer group and 8 normal liver tissue samples were collected for HE staining, then accordong to the 4 classificatin by Edmondson, we divided into different groups in the next experiment: the high differentiation of liver cancer group(n = 9), the middle differentiation of liver cancer group(n = 15), the poorly differentiation of liver cancer group(n = 6), the undifferentiated of liver cancer group(n = 0), and the control group(n = 8).2. The m RNA expressions of PI3KC3 and Beclin1 were all obviously increased in high differentiation group, middle differentiated and poorly differentiated groups compared with the control group(all P < 0.05). The m RNA expressions of PI3KC3 and Beclin1 were significantly increased in low differentiation group compared with the high differentiation and middle differentiation group(P < 0.05). There was no significant differences about m RNA expressions of PI3KC3 and Beclin1 between the middle differentiation group and high differentiation group.(P > 0.05).3. The protein expressions of PI3KC3 and Beclin1 were all obviously increased in high differentiation group, middle differentiated and poorly differentiated groups compared with the control group(all P < 0.05). The protein expressions of PI3KC3 and Beclin1 were significantly increased in low differentiation group compared with the high differentiation and middle differentiation group(P < 0.05). There was no significant differences about protein expressions of PI3KC3 and Beclin1 between the middle differentiation group and high differentiation group.(P > 0.05).4. Correlation analysis: PI3KC3 m RNA expression and Beclin1 m RNA expression were positively correlation(r=0.695 p=0.000), PI3KC3 protein expression and Beclin1 protein expression were a positive correlation(r=0.691 p=0.000).Conclusion: The exoressions of PI3KC3 and Beclin1 are higher expression in rats liver cancer tissue, the lower differentiation of liver cancer,the higher expressions of PI3KC3 and Beclin1, and the expressions of the two gene are positively related.
