| Objective: Colorectal cancer is common gastrointestinal malignancy. Autophagy is an additional adaptive mechanism by which cells recycle their own damaged organelles and proteins to generate metabolic fuel while, at the same time, preventing the accumulation of excess or defective cytoplasmic constituents. This study detected the expression pattern of autophagy markers Beclin1 and p62 in colorectal cancer and matched normal mucosa tissues, and investigated the roles of autophage in the growth, metastasis and clinical behaviour of these common tumours. Methods:The tumor and matched tissues from 32 patients with colorectal carcinomas, treated with surgery alone, were assessed immunohistochemically for Beclin1 and p62. Associations with pathological type, metastasis and clinical behaviour were sought.Results:1 The positive expression rate in colorectal tumors and normal mucosa tissue of Beclin1 protein was 71.88% and 78.94%, respectively (P>0.05). The positive expression rate in colorectal tumors and normal mucosa tissue of p62 protein was 81.25% and 47.36%, respectivey(P<0.05).2 The positive expression rate in tubular adenocarcinoma and mucinous adenocarcinoma of Beclin1 protein was 68.97% and 100%, respectively (P>0.05). The positive expression rate in tubular adenocarcinoma and mucinous adenocarcinoma of p62 protein was 82.76% and 66.67%,t respectively (P>0.05).3 The positive expression rate in without tumor infiltration serous and infiltrating of Beclin1 protein was 66.67% and 73.91%, respectively (P>0.05).The positive expression rate in without tumor infiltration serous and infiltrating of p62 protein was 66.67% and 86.96%, respectively (P>0.05).4 The positive expression rate in lymph node metastasis and without lymph node metastasis of Beclin1 protein was 75% and 68.75%, respectively (P>0.05). The positive expression rate in lymph node metastasis and without lymph node metastasis of p62 protein was 87.5% and 75%, respectively (P>0.05).5 The positive expression rate in colon and rectal of Beclin1 protein was 61.11% and 85.71%, respectively (P>0.05). The positive expression rate in colon and rectal of p62 protein was 92.30% and 73.68%, respectively (P>0.05).6 There were no correlation between Beclin1 and p62 expression in colorectal cancer (P>0.05).Conclusion:1 Autophagy activity in colorectal cancer tissue reduced.2 Autophagy is not associated with pathological types, and metastasis of colorectal cancer. |
