Objective: Observe the effects of total flavonoids from Artemisia Scoparia on anti-influenza virus, bacteriostatic, lieving cough, anti-inflammatory, anti-pyretic, And through the acute toxicity test to determine the maximum dosage. Methods:(1) Antiviral action in vivo and in vitro: Observe the effect of total flavonoids from Artemisia Scoparia on the Hep2 cells which was vaccinated by viruses. In vivo, influenza virus which rat lung adapts was given to the mice to build up the model of viral pneumonia, and then observe effects of total flavonoids from Artemisia Scoparia on the infection of lung index, mortality, and survival time in mice.(2) Bacteriostatic action: by test tube method in vitro, observe the bacteriostatic action of total flavonoids from Artemisia Scoparia on Staphylococcus aureus and Escherichia coli. In vivo, Staphylococcus aureus and Escherichia coli were inoculated to mice to build up model of bacterial Pneumonia, observe protective effects of total flavonoids from Artemisia Scoparia on the model mice.(3) Relieving cough caused by the irritation of ammonia water in mice and cough model induced by citric acid in guinea pigs were used to observe the Antitussive action.(4) Through the tests of ear conch induced by Xylene and glacial acetic acid-induced mouse peritoneal capillary inflammatory exudate model, to observe the anti-inflammatory effects of total flavonoids from Artemisia Scoparia.(5)The models of dry yeast-induced fevered rats was used to study the antipyretic effects.(6) The largest drug concentration and the maximum volume was given to the mice twice a day, observe the acute toxicity of possible responses. Results:(1) Total flavonoids from Artemisia Scoparia has inhibitory effect to cells which was vaccinated by viruses, IC50 was 6.9±0.37 and SI was 55.1±3.0. In vivo, 0.4 g/kg dose of Artemisia Scoparia flavonoids can obviously reduce the mortality of influenza virus infection mice, 0.2 g/kg and 0.4 g/kg dose can obviously prolong survival time and 0.1 g/kg, 0.2 g/kg and 0.4 g/kg dose could reduce the influenza virus infection in mice lung lesions.(2)The MIC and MBC of total flavonoids from Artemisia Scoparia to e. coli were 62.5 mg/mL and 125 mg/mL, The MIC and MBC to staphylococcus aureus were 31.25 mg/mL and 62.5 mg/mL, the results show that the total flavonoids from Artemisia Scoparia has certain protective effect in mice infected by Escherichia coli and Staphylococcus aureus.(3) The mice cough response inhibition rate was 22.2% and 35.6%,which dose of 0.2g/kg and 0.4g/kg of total flavonoids from Artemisia Scoparia; Dose of 0.1 g/kg,0.2 g/kg,0.4g/kg of total flavonoids from Artemisia Scoparia to guinea pig cough caused by citric acid inhibition rates were 32.1%, 41.3% and 49.0% respectively.(4) Total flavonoids from Artemisia Scoparia could inhibit the degree of ear conch induced by xylene and Inhibit the degree of glacial acetic acid-induced mouse peritoneal capillary inflammatory exudates model.(5)Total flavonoids from Artemisia Scoparia have obvious antipyretic effect, dose of 0.2 g/kg,0.4g/kg of total flavonoids from Artemisia Scoparia had best cooling effect that rats fever model induced after 5.5h to 7h((I.e., 2.5 to 4 hours after administration))(6)The maximum dosage to mice a day of total flavonoids from Artemisia Scoparia was 16 g/kg d, and no other acute toxicity reaction. Conclusion: The total flavonoids from Artemisia scoparia had better effect on anti-inflammatory, relieving cough and antipyretic. |
