The Experimental Study Of Tong Luo Decoction On Chronic Pelvic Inflammatory Disease Of Stagnancy Of Warm-heat Pathogen

Objective:TO explore the influence of Tong Luo Decoction on hemorhology, serum percentage of the CD4 + CD25 + regulatory T cells, cytokines and the expression of Caspase 3, the nf-kappa B protein in endometrial tissue rat with chronic pelvic inflammatory disease of Stagnancy of Warm-Heat Pathogen.Methods: The SD rats were divided into six groups, including normal group, model group, Tong Luo Decoction(10.8, 21.6, 43.2 g/kg) treated groups and Guizhi Fuling Wan(3.0 g/kg) group. Tong Luo Decoction were used daily via lavage for 15 days.CPID was induced by Phenol glue paste(by liquefied phenol, glycerin, Astragalus gum composition) in rats. At last after modeling,Tong Luo Decoction were used daily via lavage for 15 days. Whole blood viscosity, plasma viscosity, hematocrit,erythrocyte sedimentation rate equation value K were determinate by Automatic blood rheometer. Contents of Interleukin 8(IL-8), Interleukin 10(IL-10), tumor necrosis factor-α(TNF-α) were determinate by elisa. Serum percentage of CD4 + CD25 +regulatory T cells were analysed by Flow cytometric. The positive expression of cysteine proteinase-3(Caspase-3), nuclear factor-kappa B(NF-κB) were detected by Immunohistochemistry.Results:1 Compared with normol group, major pelvic tissue showed inflammatory change in Model group. Compared with model group, Tong Luo Decoction and Guizhi Fuling Wan reduced Rat pelvic tissue inflammatory change.2. Compared with the normol group, Whole blood viscosity, plasma viscosity,hematocrit, erythrocyte sedimentation rate equation value K, IL-8, TNF-α,the exprssion of Caspase-3 and NF-κB were increased obviously in model group.The level of the IL-10 were decreased obviously in model group.3. Compared with the model group, Tong Luo Decoction(21.6, 43.2 g/kg) and Guizhi Fuling Wa significantly increased the expression of IL-10 、 Caspase-3, degraded Whole blood viscosity, plasma viscosity, hematocrit, erythrocyte sedimentation rate equation value K, IL-8, TNF-α contents and the expression of NF-κB.Conclusion:Tong Luo Decoction could imorove the change of Whole blood viscosity,plasma viscosity, hematocrit, erythrocyte sedimentation rate equation value K,degrade the contents of TNF-α,IL-8, steped up the level of IL-10. By increasing the expression of Caspase-3 promote endometrial tissue inflammatory cells apoptosis.By reducing the expression of NF- κ B relieve inflammation effect.by regulating percentage the of CD4 + CD25 + Treg cells reduce the immunosuppression.Tong Luo Decoction has certain therapeutical effect effect on CPID. its mechanism may be associated with alleviation of inflammation reaction,regulation of immune function.
