Objective:Analyze the maternal and infant pregnancy outcome of Idiopathic polyhydramnios.In this experiment,EPHB4 expression in amniotic membrane and chorionic membrane of pregnant women with Idiopathic polyhydramnios and normal pregnant women was detected.To explore and analyze the correlation between EPHB4 and Idiopathic polyhydramnios.To explore the possible formation mechanism of Idiopathic polyhydramnios.Methods:1.From September 2015 to September 2019,88 pregnant women with Idiopathic polyhydramnios who had received prenatal diagnosis and delivered in the prenatal diagnosis department of the first affiliated hospital of guangxi medical university were selected as the Idiopathic polyhydramnios group,while102 pregnant women with normal amniotic fluid volume were selected as the control group.The pregnancy outcomes of the two groups were compared.2.Real-time fluorescence quantitative PCR(real-time PCR,RT-PCR)was used to detect the expression of EPHB4 m RNA in amniotic and chorionic tissues of 12 pregnant women with Idiopathic polyhydramnios(Idiopathic polyhydramnios group)and 12 normal pregnant women(control group),respectively,to analyze whether there was differential expression of EPHB4 m RNA in two groups.3.Western Blotting(WB)was used to detect the expression of EPHB4 m RNA in amniotic and chorionic tissues of 12 pregnant women with Idiopathic polyhydramnios(Idiopathic polyhydramnios group)and 12 normal pregnant women(control group),respectively,to analyze whether there was differential expression of EPHB4 protein in the two groups.Results:1.The pregnancy complications of the Idiopathic polyhydramnios group and the control group were compared.Neither group had placental abruption,umbilical cord prolapse.The results showed that the rate of premature rupture of membranes in the Idiopathic polyhydramnios group was higher than that in the control group,and the difference was statistically significant(P<0.05),while the difference between preterm delivery and fetal distress was not statistically significant(P > 0.05).2.Compared the delivery modes and incidence of birth complications of the Idiopathic polyhydramnios group and the control group,the results showed that the cesarean section rate and rate of postpartum hemorrhage of the Idiopathic polyhydramnios group was higher than that of the control group,and the difference was statistically significant(P<0.05).There was no statistically significant difference between the two groups in labor induction and forceps midwifery(P > 0.05).3.Compare the the perinatal complications of the Idiopathic polyhydramnios group and the control group,there were no neonatal and death occurred in the two groups.The results showed that the birth rate of girls,incidence of macrosomia,neonatal asphyxia rate and NICU occupancy rate of the Idiopathic polyhydramnios group were all higher than the control group,with statistically significant differences(P<0.05).4.The RT-PCR results of this experiment showed that EPHB4 m RNA was expressed in the amniotic membrane and chorionic membrane of both the Idiopathic polyhydramnios group and control group.The relative expression of EPHB4 m RNA in the Idiopathic polyhydramnios group was(0.73±0.13),while that in the control group was(0.33±0.13).The relative expression of EPHB4 m RNA in the Idiopathic polyhydramnios group was(1.77±0.44),while the relative expression of EPHB4 m RNA in the control group was(0.06±0.04).Eph B4 m RNA was highly expressed in amniotic and chorionic tissues in the Idiopathic polyhydramnios group compared with the normal group(P<0.05),with statistically significant difference.5.The Western Blotting results of this experiment showed that EPHB4 protein was expressed in the amniotic membrane and chorionic membrane of both the Idiopathic polyhydramnios group and control group.The relative expression of EPHB4 m RNA in the Idiopathic polyhydramnios group was(1.73±0.22),while that in the control group was(0.528±0.18).The relative expression of EPHB4 protein in the Idiopathic polyhydramnios group was(1.34±0.48),while the relative expression of EPHB4 m RNA in the control group was(0.58±0.15).Eph B4 protein was highly expressed in amniotic and chorionic tissues in the Idiopathic polyhydramnios group compared with the normal group(P<0.05),with statistically significant difference.Conclusion:1.Idiopathic polyhydramnios significantly affects the pregnancy outcome of mother and child,and the premature rupture rate of membranes,cesarean section rate,postpartum hemorrhage rate,the birth rate of girls,incidence of macrosomia,neonatal asphyxia rate and NICY occupancy rate were significantly increase.Patients with idiopathic polyhydramnios should be closely monitored,especially in the third trimester of pregnancy,timely termination of pregnancy can reduce maternal and infant complications.2.EPHB4 m RNA and EPHB4 protein were expressed in the amniotic and chorionic tissues of both the idiopathic polyhydramnios group and control group,and the Eph B4 m RNA and EPHB4 protein were highly expressed in amniotic and chorionic tissues in the Idiopathic polyhydramnios group compared with the normal group.
