Objective Using rabbits as experimental object, muscle-thickness abdominal wall defect models were established, with the method of packet control, to study the postoperative inflammation of tension-free herniorrhaphy, and the application of Aescuven forte to alleviate the postoperative inflammation of tension-free herniorrhaphy.Objective Using rabbits as experimental object, muscle-thickness abdominal wall defect models were established, with the method of packet control, to study the postoperative inflammation of tension-free herniorrhaphy, and the application of Aescuven forte to alleviate the postoperative inflammation of tension-free herniorrhaphy.Methods Randomly selected 18 rabbits of clean grade, from the animal laboratory of Anhui medical university, muscle-thickness abdominal wall defect models were established, and randomly assigned to one of three groups:repaired without any mesh(negative control group, A), repaired by polypropylene mesh group(control group, B), and the group was repaired by polypropylene mesh after oral Aescuven forte(experimental group, C):2 days before the operation for medicine, daily 100mg/kg. Acquisition biood from ear vein of the rabbit at the time points of preoperative and after 1,4,7,20 d, Centrifugal serum, and the expression of tumor necrosis factor(TNF)-a and interleukin(IL)-β was detected simultaneously by using radioimmunoassay(RIA) and on the postoperative day 1,4,7 and 20 in each group. Using the variance analysis to compare within group, and the q test for comparison between groups; kill the rabbits with the air needle after 20d, observe and score the incision tissue edema,using poncirus related inspection to analysis swelling scores; take the surrounding tissue adhesion after the rabbits were killed, and immersed into 10%Formaldehyde Solution, according to conventional methods for specimens for HE staining, and observe the pathological change with light microscope.Results There was no death in 3 groups, abdominal incision infection occurred in 1 cases in B group. The score of incision edema of the rabbits in the experience group(n=6) was 3,and the control group(n=6) was 13,the difference was statistically significant between the 2 groups(P<0.05);The preoperative differences of the levels of TNF-a and IL-1β in 3 groups showed no significance[TNF-α was (1.35±0.14)、 (1.19±0.12)、(1.11±0.32)μg/L respectively and IL-1β was (0.052±0.011). (0.053±0.022)、(0.073±0.024)μg/L respectively, P>0.05)];4 and 7 days after the operation, the levels of TNF-a and IL-1β in the control group were significantly increased as compared with the blank group and the experience group[control group TNF-awas (3.40±0.59)μg/L and(5.5±0.8)μg/L,IL-1βwas(0.35±0.161)μg/L and(0.655±0.132) μg/L, blank group TNF-αwas (1.48±0.29) μg/L and (1.6±0.4) μg/L, IL-1β was (0.113±0.015) μg/L and (0.120±0.015) μg/L, experience group TNF-α was (2.07±0.53) μg/L and (2.4±0.6) μg/L, IL-1β was (0.237±0.079) μg/L and (0.237±0.079) μg/L, P<0.05]. Compared with the blank group, inflammatory cytokines were significantly increased in experimental group and control group, the biggest difference was at 1 weeks after operation, however, the increased levels of experimental group was significantly reduced than the control group, it was showed that there is still a certain inflammatory response after taking Aescuven forte, Aescuven forte can alleviate the postoperative inflammation of tension-free herniorrhaphy and worthy of popularization and application.Results There was no death in 3 groups, abdominal incision infection occurred in 1 cases in B group. So Aescuven forte can use in clinical application, and effectively reduce the postoperative complications caused by inflammatory reaction after the tension-free herniorrhaphy.
