Objective To establish a stable cell line with stable expression of PES1 sh RNA in three kinds of tongue cancer cells,Tca8113, SCC6 and SCC15.Study on the effect of knocking down PES1 gene on the growth of tongue cancer cells, and further study the regulation of PES1 gene in tongue cancer cell cycle and effect on invasion and migration of tongue cancer cells.Methods Using virus packaging plasmids to transfect 293 T cells to obtain the virus of low PES1 gene,which were used to infect tongue cancer cells. Then the stable clones of cells stably expressing low PES1 gene were screened.The expression level of PES1 protein was detected by Western Blot.The effect of PES1 gene on the growth of tongue cancer cells was detected by cell growth curve when the effect of the cell cycle of tongue cancer cells was detected by the cell cycle experiment.Migration rate of tongue cancer cells was measured by wound healing assay.The invasion ability of tongue cancer cells was detected by the transwell assay.Results(1) We successfully obtained the tongue cancer cell line stably expressing PES1 sh RNA gene.(2)The growth rate of tongue cancer cells with knockdown of PES1 gene was lower than the control group.(3)Knockdown of PES1 gene can cause cell cycle arrest of tongue cancer cells in G1 phase, which can inhibit the expression of Cyclin D1.(4)The migration of tongue cancer cells with knockdown of PES1 gene was lower than the control group.(5)The invasion ability of tongue cancer cells with knockdown of PES1 gene was lower than the control group.Conclusion Knockdown of PES1 gene in tongue cancer cells can inhibit the growth of tongue carcinoma cells with the cancer cell cycle arrested in G1 phase, and the expression of cyclin D1 inhibited.Knockdown of PES1 gene in tongue cancer cells can inhibit the migration and invasion ability of tongue cancer cells.PES1 may become the target of cancer gene therapy.
