Objective:1.To explore the effect on cell cycle of tongue squamous carcinoma cell line Tca8113after different doses of heavy ion beam and X-ray irradiation.2.To discuss the mechanism of tongue squamous carcinoma radiotherapy by heavyionbeam,to make sure the effectiveness and safety are valid, and to provide prac tical evidence for clinical treatment.Materials and Methods:The cell line Tca8113was kindly provided by Professor Jia Jun from Wuhan University,cryopreserved and stored by Professor Feng Zhenghu from Northwest University for Nationalities. The cells were cultivated for thefollow-up experiments. The cells were divided into2groups-experiment group(irradiated by heavy ion beam) and control group(irradiated by X-ray):each group was divided into3subgroups of different doses-0Gy,1Gy,4Gy. The cells of each group were collected after the irradiation for12h,24h,36h and48h.The heavy ion beam offered by HIRFL was12C ion beam(250MeV/μ70KeV/μm) at a dose rate of0.5Gy/min. The dose rate of X-ray was0.3Gy/min, and the energy was50kVp.The cell cycle arrest and the expression of VEGF were detected by flow cytometry and Southernblot,respectively.Results:1.Cell cycle study result showed that, study group4Gy appeared G2/M phase arr est obviously at36h and48h (p<0.01). Control group4Gy appeared G2/M phase arres t at24h and36h (p<0.05).2.In situ hybridization implied that, the positive expression rate of VEGF for stu dy group was84.7%for OGy, and21%for4Gy group at8h, for control group, it was84%for OGy, and55%for4Gy at8h. Although both of them were4Gy at8h, there were still obvious difference between them (p<0.05)..Conclusion:1.The cell division inhibition effect for tongue squamous carcinoma cells(Tca8113) was more obvious after heave ion irradiation than X-ray irradiation. Heavy ion irr-a diation was a better cure for tongue squamous cell carcinoma than X-ray irradiation2.VEGF of Tca8113cell was much lower after heavy ion irradiation than X-ray i rradiation, they have staitistical differences. The inhibition effect to canser of heavy i con irradiation was much obvious than X-ray irradiation, Heavy icon irradiation appea rd to be a safer solution for clinical application.
