Objective To investigate the expression of intercellular adhesion molecule-1(ICAM-1) and vascular cell adhesion molecule-1(VCAM-1) in the serum and tumor tissue of colorectal carcinoma,and detect their function in the occurrence and development of colorectal carcinoma.MethodsThe preoperative serum of 78 patients with colorectal carcinoma served as serum experimental group, the serum of 40 normal persons as serum control group.Enzyme-linked immunosorbent assay (ELISA) method was used to detect the expressions of ICAM-1 and VCAM-1 in the serum. The postoperative tumorous tissues of 78 patients with colorectal carcinoma served as tissular experimental group, and the postoperative para-tumorous tissues of 78 patients with colorectal carcinoma served as tissular control group. Immunohistochemistry (IHC) method was used to detect the expressions of ICAM-1 and VCAM-1 in the tissue. Analysis the relationship between the expressions of ICAM-1 and VCAM-1 and the occurrence and development of the colorectal carcinoma.The data were statistical analyzed by SPSS 13.0.Resultsl.The expression levels of serum ICAM-1 Serum concentration of ICAM-1 in patients with colorectal carcinoma (356.6±44.3) μg/L was significantly higher than the health people (237.7±34.5) μg/L, there was statistically significant (t=14.86,P<0.01).2.Expression of tissue ICAM-1 The expression rate of ICAM-1 in the tissular experimental group (62.8%) was significantly higher than that of the tissular contral group (19.2%).There was significant difference between the high expression of ICAM-1 in colorectal carcinoma and the low expression in normal mucosa (χ2=30.64,P<0.01).The expression of ICAM-1 in colorectal carcinoma were associated with lymph node metastasis and Dukes stage (P<0.01),but had nothing to do with sex, age, tumor location and histological grades (all P>0.05).Positive lymph node metastasis group was significantly higher than that of negative lymph node metastasis group (P<0.01).Dukes stage C/D group was significantly higher than that of Dukes stage A/B group (P<0.01)3.Expression correlation of serum ICAM-1 and tissue ICAM-1 in colorectal carcinoma Serum concentration of ICAM-1 with positive tissual group (392.5±42.4) μg/L was significantly higher than the negative tissual group (295.9±47.5)μg/L, there was statistically significant (t=9.28,P<0.01).Tissue ICAM-1 and serum ICAM-1 expression in colorectal carcinoma was positively correlated.4.The expression levels of serum VCAM-1 Serum concentration of VCAM-1 in patients with colorectal carcinoma (898.6±51.2) μg/L was significantly higher than the health people (457.3±35.7)μg/L, there was statistically significant (t=48.64,P<0.01).5.Expression of tissue VCAM-1 The expression rate of VCAM-1 in the tissular experimental group (78.2%) was significantly higher than that of the tissular contral group (14.1%).There was significant difference between the high expression of VCAM-1 in colorectal carcinoma and the low expression in normal mucosa (χ2=64.48,P=0.000).The expression of VCAM-1 in colorectal carcinoma were associated with lymph node metastasis and Dukes stage (P<0.01),but not with sex, age, tumor location and histological grades (all P>0.05).Positive lymph node metastasis group was significantly higher than that of negative lymph node metastasis group (P <0.01).Dukes stage C/D group was significantly higher than that of Dukes stage A/B group (P<0.01)6.Expression correlation of serum VCAM-1 and tissue VCAM-1 in colorectal carcinoma Serum concentration of VCAM-1 with positive tissual group (976.3±54.5) μg/L was significantly higher than the negative tissual group (619.8±39.4) μg/L, there was statistically significant(t=25.10,P<0.01).Tissue VCAM-1 and serum VCAM-1 expression in colorectal carcinoma was positively correlated.7.Expression correlation of serum ICAM-1 and serum VCAM-1 of colorectal carcinoma Serum ICAM-1 and serum VCAM-1 expression in colorectal carcinoma was positively correlated (χ2=7.052, r=0.301, P<0.01)8.Expression correlation of tissue ICAM-1 and tissue VCAM-1 of colorectal carcinoma Tissue ICAM-1 and tissue VCAM-1 expression in colorectal carcinoma was positively correlated (χ2=7.052, r=0.301, P<0.01)Conclusion1. Serum ICAM-1 and serum VCAM-1 high expressed in colorectal carcinoma speculated that they could be a new tumor marker for colorectal cancer to improve the rate of diagnosis.2. Tissue ICAM-1 and tissue VCAM-1 high expressed in colorectal carcinoma, and their expression were related with the lymphatic metastasis and staging of colorectal carcinoma,speculated that they may play an important role in evaluation of malignant degree and judging the progress as well as the therapy of colorectal cancer.3. Serum ICAM-1 and serum VCAM-1 expression in colorectal carcinoma was positively correlated,speculated that they combine detection may provide a new tumor marker for the diagnosis of colorectal carcinoma.4. Tissue ICAM-1 and tissue VCAM-1 expression in colorectal carcinoma was positively correlated,through one or a variety of ways to participate both development and metastasis of colorectal carcinoma.
