Experimental Study On Health Functions Of Three Kinds Traditional Chinese Medicines

bjective: To investigate the lipid-lowering, laxative effect of Tongrun granules, the effect and mechanism of Compound Shenmai capsule on anti-fatigue action, as well as the effect of improving sleep of Anshui granules.Method: 1. Hypolipidemic effect: To establish hypercholesterolemic rat model in a high-fat diet, then some indexs were tested that levels of the total cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL-C), low density lipoprotein(LDL-C), Alanine aminotransferase(ALT) and Aspartate transaminase(AST) in those rat’s blood, and calculated LDL-C/HDL-C ratio. 2. The laxative Function: Gavage KM mice with Loperamide + 5% Atropine to establish constipation model, then measure ink propulsion rate in intestinal within 0.5 hours after gavaged tongrun granules, record grains and weight of bowel within 6 hours. 3. The anti-fatigue effect: Healthy male KM mice were given Compound Shenmai capsule daily for 30 days. Then all animals were used in experiments to test the loading swimming time; the Content of hepatic glycogen; The Urea content and The Creatine kinase(CK) activity; the Content of blood Lactate,and calculate the Area under the curve of it. 4. Improve sleep function: Healthy female KM mice were gavaged Anshui granule daliy for 30 days, observe the direct sleep action, All mices were used after 30 days in experiments, to determinate the sleeping time extension of mices when they were hypnotized by Pelltobarbitalum Natricum, and to determinate the numbers of sleeping mice when they were hypnotized by subthreshold dose of Pelltobarbitalum Natricum. last to determinate sleep latency when they were hypnotized by Barbitone sodium.Results: 1. Hypolipidemic effect : Compared with the model group, serum cholesterol(TC) and low-density lipoprotein(LDL-C) in rats all were significantly reduced by two dose groups of Tongrun granule(6.2,12.4g native drug/kg)(p<0.05 或 p<0.01);but the level of triglyceride(TG) among all groups rats had no significant difference(p>0.05), high-density lipoprotein(HDL-C) levels lipoprotein(HDL-C) levels were increased by three dose groups of Tongrun granule(3.1,6.2,12.4g native drug/kg)(p<0.05或p<0.01). So LDL-C/HDL-Cratio were reduced(p<0.05或p<0.01). Alanine aminotransferase(ALT) and aspartate transaminase(AST) levels can also be significantly reduced(p<0.05 或 p<0.01). 2. The laxative Function : compared with the model group, the ink propulsion rate of 0.5 hours in small intestine of thoes mices can be significantly improved(p<0.05 或 p<0.01), and weight and grain number of feces of mice within 6 hours can be increased after they were gavaged three doses of Tongrun granule for seven days(p<0.05 或p<0.01). 3. The anti-fatigue effect and mechanism study of Shenmai Compound Capsules: the loading swimming time could be significantly prolonged by two dose groups of Compound Shenmai Capsule(5.34,10.68 g native drug/kg)(p<0.05 或 p<0.01), and the level of serum urea and the activity of creatine kinase in mices’ blood could be significantly reduced by three dose groups of Compound Shenmai capsule(2.67,5.34,10.68 g native drug/kg)(p<0.05 或 p<0.01)who could’not only significantly increase the content of liver glycogen, but also reduce the content of lactate in mices’ blood and the area under the curve of its(p<0.05 或 p<0.01).4. Improve sleep funvtion: compared with the control group, Anshui granule 7.0,14.0g native drug/kg dose groups could significantly prolong sleep latency of mice who were hypnotized by pentobarbital sodium(p<0.05 或 p<0.01), significantly increased the number of mices got to sleep when they were hypnotized by subthreshold dose of pentobarbital sodium(p<0.05 或 p<0.01)and Anshui granule 14.0,7.0,3.5 g native drug / kg three dose groups could’nt directly hypnotic mices(p> 0.05). Anshui granule 14.0,7.0,3.5 g native drug/kg three dose groups extend the trend to the sleep time of mice hypnotized by pentobarbital sodium, but compared with the control group, there was not statistically significant difference(p> 0.05).Conclusion: 1.Test results suggste that Tongrun granule has the function of lipid-lowering. It maybe prevent the development of hypercholesterolemia throug the mechanism of reducing the content of total serum cholesterol and the low-density lipoprotein levels, and increasing high-density lipoprotein levels; In addition, it has certain effect on protecting liver injury. 2.Tongrun granule has significant cathartic action. 3.Compound Shenmai capsule has obvious anti fatigue effect, may be through decreasing the content of serum urea and the activity of creatine kinase, improveing liver glycogen storage, delaying the generation of blood lactic acid and accelerateing the removal of it. 4.Anshui granule has the function of improving sleep.
