Background:With the social pressure increasing,and unhealthy lifestyle has led to the decline in people’s health state,resulting in different kinds of physical and mental discomfort,however,modern medicine can not diagnose it as any kinds of disease,and just call it sub-health status.Sub-health status has seriously influenced health condition of people.Fatigue is the most common symptoms of sub-health,prevention and treatment of fatigue are the key of sub-health research.Objective:By this study,we tend to know the effect of Weikang granule on intervening sub-health fatigue mice,discuss characteristics of serum metabolomics,and provide basis for the prevention and treatment of sub-health fatigue Weikang granule.Method:50 male 4 week-old SPF level Kunming mice were used in this study.Bef-ore the study,all of the mice were quarantined for 1 week,with adaptive feeding for one week,swimming training for 3 weeks,3 times per week,5 min each time.After that,exhausted swimming by bearing 7%of mice body weight was given,forcing them to swim until exhausted,and recorded the swimming time.Then all the mice were stratified divided into blank control group,model control group,Weikang granule high-dose group,medium-dose group,low-dose group based on their exhausted swimming time and body weight.The blank control group was fed normally,while the other four groups used the modeling method based on previous study(The mice were forced into a water cage with a depth of 0.8cm,18h/day,for consecutively 7 days,1 mouse/cage).During the modeling,mice of Weikang granule groups were given high,medium,low-dose of Weikang granule by gavage,once per day,while the blank control group and model control group were given equal volume of pure water by gavage.Body weight,mental status,hair color were observed.Exhaustive swimming was used to measure the degree of fatigue after the modeling,and apex cordis,gastrocnemius and liver were observed by histopathology and electron microscopy,serum were detected for 1H NMR metabonomics research.Result:1.Compared with the blank controlg group,the body weight of other groups were significantly decreased(P<0.05),the mice had poor mental status,less glossy hair color and tended to irritable,exhausted swimming time decreased significantly(P<0.05).Compared with the model control group,there was no significant difference in body weight of the Weikang granule groups(P>0.05),the mental status was better and the hair color was more shiny.Among them,the exhausted swimming time of the high-dose group was significantly different from that of the model group(P<0.05).2.There were no pathological changes noted in histopathology in each group.The results of electron microscopy indicated that mitochondria was damaged and autophagosome were formed in the heart,gastrocnemius and liver tissue of the model group,and the damage degree of mitochondria in the Weikang granule groups was obviously relieved,and the number of autophagosomes decreased.3.Serum metabolomics results indicated that,compared with blank control group,expression of isoleucine,valine,alanine,leucine,lysine,ketoisocaproate,glutamate,pyruvate,glutathione,histidine were significantly increased,while dimethylamine,glucose and linolenic acid were obviously decreased in model control group.Compared with model control group,expresion of isoleucine,valine,alanine,leucine,lysine,citrulline,ketoisocaproate,acetoacetate,proline,and glycine were significantly decreased,and glucose was increased differently after intervented by Weikang granule.While isoleucine,valine,alanine,leucine,ketoisocaproate,and glucose were readjust after invervention,suggesting that these 7 metabolites maybe the special metabolites of Weikang granule on interventing subhealth,they invoved in carbohydrate metabolism,lipid metabolism,amino acid metabolism,three tricarboxylic acid cycle energy metabolism pathway.Conclusion:Weikang granule can prevent and treat subhealth fatigue by reducing mitochondrial damage and improving energy metabolism.
