Objective: Studying on the wind drugs(Fangfeng、Baizhi) lowering blood pressure in hyperactivity type renal hypertensive rats explores the antihypertensive synergistic action and mechanism of the wind drugs in hypertension treatment, and provides scientific basis for the wind drugs treating hypertension. Methods: The screening of male SD rats were 100, 12 were divided randomly into normal group, 76 were into surgery group and 12 were into sham group. The surgery group were ligated the left renal artery to build renal hypertensive rats. There were 55 successful rats who fed with aconite soup for 4 weeks after 2 weeks operatively to build hyperactivity type renal hypertensive rats(53 were successful finally). The model successful rats were screened out of 50 randomly to divide into model group,captopril group, wind drugs group, Tianma Gouteng decoction group and wind drugs+Tianma Gouteng decoction group(referred to comprehensive group). Each group had 10 rats. The rats were fed the corresponding drugs, once a day. The normal group, sham group and model group were given the same volume of physiological saline. After 4 weeks, the rats were took the blood of abdominal aortic to prepare serum. The chemical colorimetry was used to determinate serum NO, and ELISA method was used to determinate serum ET, TXB2, 6-K-PGF1 a. Taking the thoracic aorta and the right renal artery, fastened, entrapped with paraffin, section, HE staining, then observing the change of vascular membrane and measuring vascular membrane area with light microscopy. Immunohistochemistry was used to detect the expression of AT1 R in the thoracic aorta. Results: 1. Observation of the general situation and behavior: The normal group and sham group rats were clean and glossy, water intake normal, gentle, no shorter rotation time. The rats in model group were gradually yellow, tired lying, increased water intake, rotation time is shortened. Rats were irascible and even bite each other after fed aconite soup. The mental state was improving in each drug group rats, the gloss coat were gradually increasing, and dociled temperament during the post-treatment. 2. The impact of drugs on the rats blood pressure in hyperactivity type renal hypertensive rats: ① The basis blood pressure: There is no statistical difference among the each group. ② The blood pressure after 2 weeks and 6 weeks of the surgery: There is no statistical difference between the sham group and normal group; compared with the sham group, the model group were obvious increase(P<0.05); there is no statistical difference among the each drug group. ③ The blood pressure after 4 weeks of treatment: There is no statistical difference between the sham group and normal group; compared with the sham group, the model group were obvious increase(P<0.05); compared with the model group, the wind drugs group were not obvious decrease and no statistical difference, the other drug groups were obvious decrease(P<0.05); and compared with the wind drugs group and Tianma Gouteng decoction group, the comprehensive group were obvious decrease(P<0.05), there is no statistical difference between the Tianma Gouteng decoction group and captopril group. 3. The impact of drugs on vasoactive substances of hyperactivity type renal hypertensive rats: ① The serum ET and TXB2: There is no statistical difference between the sham group and normal group; Compared with the sham group, the model group were obvious increase(P<0.05); compared with the model group, the wind drugs group were not obvious decrease and no statistical difference, the other drug groups were obvious decrease(P<0.05); and compared with the wind drugs group and Tianma Gouteng decoction group, the comprehensive group were obvious decrease(P<0.05), there is no statistical difference between the Tianma Gouteng decoction group and captopril group. ② The serum NO and 6-K-PGF1a: There is no statistical difference between the sham group and normal group; compared with the sham group, the model group were obvious decrease(P<0.05); compared with the model group, the wind drugs group were not obvious increase and no statistical difference, the other drug groups were obvious increase(P<0.05); and compared with the wind drugs group and Tianma Gouteng decoction group, the comprehensive group were obvious increase(P<0.05), there is no statistical difference between the Tianma Gouteng decoction group and captopril group. 4. The impact of drugs on AT1 R expression in the thoracic aorta of hyperactivity type renal hypertensive rats: AT1 R expression was brown granules and mainly located in the thoracic aorta smooth muscle under the light microscope. AT1 R was a small amount of expression in the normal group and sham group; there is no statistical difference between them. AT1 R was an amount of expression in the model group, compared with the sham group, the model group were obvious increase(P<0.05); compared with the model group, the wind drugs group were not obvious decrease and no statistical difference, the other drug groups were obvious decrease(P<0.05); and compared with the wind drugs group and Tianma Gouteng decoction group, the comprehensive group were obvious decrease(P<0.05), there is no statistical difference between the Tianma Gouteng decoction group and captopril group. 5. The impact of drugs on the membrane change of rat thoracic aorta and right renal artery and membrane area of hyperactivity type renal hypertensive rats: Under the light microscope, compared with the normal group, the sham group were no pathological change and membrane area had no statistical difference; compared with the sham group, in the model group, thoracic aortic and right renal artery medial smooth muscle cells and elastic fibers were hyperplasia, disorganized, membrane were thickness, and elastic fibers were fracture in thoracic aortic; the membrane area were obvious increase(P<0.05); compared with the model group, the proliferation and membrane area in the wind drugs group were not obvious decrease and no statistical difference, the other drug groups were obvious decrease(P<0.05); and compared with the wind drugs group and Tianma Gouteng decoction group, the comprehensive group were obvious decrease(P<0.05); there is no statistical difference between the Tianma Gouteng decoction group and captopril group. Conclusion: 1. Through partial ligating left renal artery and irrigating aconite soup, the model rats with hyperactivity type renal hypertensive rats were preparated successfully. 2. The wind drugs combining with Tianma Gouteng decoction had more obviously antihypertensive effect than the single Tianma Gouteng decoction on hyperactivity type renal hypertensive rats, the wind drugs can enhance the effect of antihypertension in the Tianma Gouteng decoction. 3. Antihypertensive mechanism may be related to the following factors: ① Reduce the formation of vasoconstrictor ET, reduce the intensity of vascular contraction caused by ET; and weak the positive effect of ET, and increase vasodilator NO production, then dilate blood vessels, low blood pressure, and antagonize the vascular contraction effect of ET. ② Stable endothelial function and promote TXB2/6-K-PGF1 a levels normal, keep blood circulation normal, prevent vasospasm and thrombosis from formation. ③ Reduce the AT1 R expression in vascular smooth muscle, inhibit vasoconstriction and prevent vascular smooth muscle cells from roliferating and migrating, thereby inhibit vascular reconstruction.
