Abies fargesii and Picea asperata are the main species in natural spruce-fir forest in Qinba Mountain. Because of the abundant resources and large growth increment, it is necessary and urgent for using the tending slashes. In this study, the kinds of flavonoids in Abies fargesii and Picea asperat were studied. The ultrasonic extraction process of flavonoids form Abies fargesii and Picea asperata was optimized through response surface methodology. The antioxidant activity was analyzed with DPPH and ABTS methods..The results showed that:(1) The kinds of flavonoids in Abies fargeii and Picea asperata were different. The color results showed that, there were chalcones and flavonols in the extraction solution of Abies fargesii and Picea asperata.The dihydro flavonoids was found in the extraction of Abies fargesii, while the other one was not. In the Picea asperata, autones was not exsit, while another one was uncertain.(2) In the extraction process of flavonoids from Abies fargesii, ultrasonic temperature, ultrasonic time, ultrasonic power and the concentration of ethanol had significant effect on the yield. And the factors influences were as follows: ethanol concentration>temperature>power>time. The optimizationed conditions were ethanol concentration 55%, temperature 66℃, power 300 W, time 40 min. The maximum yield was 4.817%.(3) In the extraction process of flavonoids from Picea asperata, ultrasonic temperature, time, power and the concentration of ethanol had significant effect on the yield. The factors influences were as follows: time>temperature>ethanol concentration>power. The optimizationed conditions were ethanol concentration 48%, temperature 56℃, ultrasonic power 300 W, time 41 min. The maximum yield was 4.745%(4) Abies fargesii and Picea asperata had antioxidant ability. But they were weaker than Vc and quercetin. The ability to clear DPPH? of Abies fargesii is stronger than Picea asperata, and the IC50 were 20.315 μg/mL and 21.979g/mL respectively. In the test of ABTS?, the result of Picea asperata was higher than Abies fargesii, similar with quercetin, weaker than Vc, and the IC50 were 11.836 μg/mL and 6.975μg/mL respectively.
