Objective:This study is to investigate the pulmonary surfactant protein A2 (SP-A2) haplotype in premature infants relationship with respiratory distress syndrome (RDS), and understand the distribution of SP-A2 haplotypes in the population of Guangxi in the local area.Methods:Using population base and case-control study cohort; We selected 80 cases of preterm children with RDS and 80 cases of non-RDS in preterm children for the study, and the subjects that selected 80 infant of full-term delivery were used as healthy infants crowd; Intravenous or cord anticoagulant of the object of study was collected, use DNA extraction kit to extract genomic DNA; SP-A2 gene of four single nucleotide polymorphisms (rs1059046, rsl7886395, rs1965707, rs 1965708) were genotyped by using TaqMan(?) real-time quantitative polymerase chain reaction (polymerase chain reaction, PCR) method;Each SNP was randomly selected from 45 cases were detected by polymerase chain reaction and DNA sequencing technology to verify the TaqMan real-time PCR genotyping results.Results:1. Basic information about case-control study, there was no difference among the case and control in gestational age, weight and sex.2. Genotypes about four sites of SP-A2 gene (rs1059046, rsl7886395, rs 1965707, rs 1965708) in term group are in line with Hardy-Weinberg equilibrium (P>0.05).; full-term children expressed mainly haplotype for 1A0,1A5,1A1, frequencies were 54.2%,21%,9.7%.3. Comparing the premature case group and the control group, each haplotype frequency distribution were not statistically different (P> 0.05).4. In 32 weeks of gestational age for the sector,compareing case group with control group,the SP-A2 haplotype frequency distribution were not different in the two groups (P> 0.05).Conclusion:1. Only SP-A2 does not contribute to the susceptibility for preterm RDS.2. SP-A2 haplotype distribution in a local Guangxi population shows ethnic characteristics to some extent.
