Now with the development of social science and technology, medical standardsare ever-changing. The emerging diagnostic techniques provide more accurate in-formations for diseases cure. More and more diseases caused by genes are foundby people, and we pay more attention to research on genetic diseases. In general,haplotypes are often considered to be the rich carriers of linkage informations ofdiferent SNPs, so the estimation of haplotype frequency is great valuable. Peopleare mostly concentrated in the haplotypes of multiple sites which have two allelesin each locus, and lots of diseases controlled by multiple alleles have been foundthrough the clinical diagnosis, so we shoud pay more emphasis on the research ofmulti-allelic haplotype.It is an important matter to research the haplotype frequencies of multiplealleles. This thesis embarks from reducing workload, cutting research cost andshortening research period to estimate the haplotype frequencies of multiple allelesthrough the EM algorithm, which uses the method of DNA pooling. In the thesis,each DNA pooling has two specimens. Its primary study is the fact that haplotypehas two sites and each site has three alleles. Through observation data obtained byobserving DNA pooling, we use the idea of EM algorithm to estimate the haplotypefrequencies of multiple alleles, and conduct the simulation by statistic software andhave an analysis in the end.
