| Objective:Study the syndrome factor distribution of advanced gastric cancer; Detect the methylation status of the promoter region in tumor suppressor gene ING1 of gastric carcinoma then compare it with the adjacent tissues; investigate the relationship between syndrome factor distribution of advanced gastric cancer and methylation status of tumor suppressor gene ING1; clarify the connection of pathogenesis in gastric cancer between traditional Chinese medicine and western medicine.Methods:To get the main syndrome elements of disease location and disease nature in advanced gastric cancer, standardized collect four diagnostic information of 60 cases of advanced gastric cancer patients and analyses the collected data. Meanwhile, collect 60 cases cancer tissues and adjacent tissues (more than 5cm away from neoplastic foci)of advanced gastric cancer patients, and use the method of methylation specific PCR (Methylation-specific PCR, MSP) to detect the methylation status of the suppressor gene ING1 in cancer tissues and adjacent tissues.Results:1.Age distribution characteristics:mainly in 60-69 years old, more men than women, the rate of male to female is 3:1.2.Syndrome factor distribution characteristics:the main syndrome elements of disease location is in the stomach, followed by the spleen, liver, and kidney; the main nature of disease is qi deficiency, and followed by wet, blood stasis, phlegm,qi stagnation.3. The methylation rate of tumor suppressor gene ING1 in gastric cancer is 71.67% (43/60), while the corresponding adjacent tissues is 25.00%(15/60). And the methylation rate of tumor suppressor gene ING1 in gastric cancer is higher than the adjacent tissues. The difference between above analysis has the statistical significance(P<0.01). While,there is not statistical difference between methylation status of suppressor gene ING1 and patient age, gender, tumor tage, lymph node metastasis and tumor location (P> 0.05).4.The syndrome elements "stomach", "qi deficiency", "blood stasis", "phlegm" is related to the methylation status of tumor suppressor gene ING1(P<0.05). But the other syndrome elements is not statistical difference related to the methylation status of tumor suppressor gene INGl(P>0.05).Conclusion:1.The main syndrome elements of disease location in advanced gastric cancer is stomach, and followed by the spleen, liver, and kidney; the main syndrome elements of disease nature is qi deficiency, followed by wet, blood stasis, phlegm,qi stagnation.The manifestation of pathogenesis in gastric cancer is deficient root with overdo superficial. The deficient root is mainly refer to qi deficiency and overdo superficial is mainly refer to wet, blood stasis, phlegm,qi stagnation.The clinical treatment of the disease can be done on the basis of strengthening healthy energy, together with removing wet, removing blood stasis, reducing phlegm, regulating vital energy and so on.2. The methylation rate of tumor suppressor gene ING1 in gastric cancer tissue is higher than the adjacent tissues, suggesting that methylation of tumor suppressor gene ING1 is related to the occurrence and development of gastric cancer.3. Syndrome elements characteristics of advanced gastric cancer is related to methylation status of suppressor gene ING1. There is connection of pathogenesis in gastric cancer between traditional Chinese medicine and western medicine.4.The syndrome elements "stomach", "qi deficiency", "blood stasis", "phlegm" is related to the methylation status of tumor suppressor gene ING1(P<0.05).
