| Objective:To investigate the expression of IL-34 and the effect of rosuvastatin on IL-34 and atherosclerosis of atherosclerotic rat aorta.Method:36 male SD rats adaptive fed for 1 week, were randomly divided into control group(n=12) and experimental group(n=24). Rats in the experimental group were high cholesterol fed for 4 weeks, then randomly divided into two groups: model group(n=12) and rosuvastatin group(n=12). NS and rosuvastatin(5mg/kg/d) were prescribed separately for 4weeks. At the end of experimental period, the serum levels of TC and TG were detected. The pathological changes of aorta was evaluated by hematoxylin-eosin staining. The expression of IL-34 of aorta was observed by immunohistochemical techniques. The expression of IL-34 mRNA of aorta was measured by reverse transcription polymerase chain reaction(RT-PCR).Results:1.Blood lipid: The level of TC in model group and rosuvastatin group were both decreased compared with control group(P<0.05). The TC leve in rosuvastatin group was significantly lower than the model group(P<0.05). There were no significant differences of TG level among the three groups(P>0.05).2.Histopathological changes in aorta: The aortic wall structure of each layer was relatively intact, no lipid calcification and deposition were observed in control group. In model group, smooth muscle cells arranged irregularly, and the swollen and deformation were seen in endothelia cells and subendothelial edema. In the thicken aortic wall, a large number of lipid deposition and formation of atherosclerosis were observed. A significant regression of atherosclerosis changes was found in rosuvasatin group when compared with the model group.3.Expression of IL-34 protein in aorta: A small amount expression of IL-34 protein was observed in aorta in control group. An increase of IL-34 was observed in both model group and rosuvasatin group when compared with control group(P<0.05). A decrease of IL-34 was found in rosuvastatin group as compared with model group(P<0.05).4.Expression of IL-34 mRNA in aorta: In model group, an increase of the expression of IL-34 mRNA was observed compared with control group and rosuvastatin group. In rosuvastatin group, an increase of the expression of IL-34 mRNA was found when compared with control group,but a decrease was observed as compared with model group.Conclusion:1. The expression of IL-34 in atherosclerotic rats increased.2. Rosuvastatin may place a role to improve atherosclerosis by reducing the expression of IL-34. |
