Objective To investigate the effects of rosuvastatin on expressions of B cellleukemia/lymphoma-2(Bcl-2) and Bcl-2-associated X protein(Bax) in aortas of Wistarrats with hyperhomocysteinemia(Hhcy).Methods33healthy male Wistar rats,6weeks old, weight145±10g, werepurchased from the Experimental Animal Center of Lanzhou University(Certificate of Conformity: SCXK (Gan)2009-0004). Specific pathogen-free(Specific pathogen free, SPFWistar rats) feeding level, natural lighting, timingventilation, a clean environment, greenhouse19-23°C, relative humidity of70%-80%packet of animal models of adaptive fed for two weeks after33according toa completely randomized design, randomizeddivided into three groups: the controlgroup of rats fed with normal diet11, the same dose of drinking water gavage weresacrificed20weeks the; Hhcy group11: rats fed with normal diet whileDL-methionine dissolved in saline, given orally (1g/kg·d),20weeks weresacrificed; rosuvastatin group11: rats fed with normal diet, the DL-methioninedissolved in normal saline for20weeks, and gavage DL-methionine, while addinggavagerosuvastatin statin (10mg/kg d) orally, the rats were killed. cardiac bloodwas collected, and collection of rat aorta specimens..Serum triglyceride (TG),total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) and plasmahomocysteine (Hcy) concentration were detected by ADVIA2400automaticbiochemical analyzer, pathological changes of aorta were observed by HE stainingand Bcl-2and Bax expressions of aortic tissues were detected by immunohistochemistry and RT-qPCR detection.Results Plasma Hcy concentration was lower in rosuvastatin group than inHhcy group,(10.47±0.07) μmol/L vs.(85.38±0.2) μmol/L, with statisticallysignificant differences (P<0.05).HE staining aortic atherosclerosis damage:Aortic injury was less severe in rosuvastatin group than in Hhcy group.Immunohistochemistry demonstrated: control group, no significant changes inAortic tissue immunohistochemistry showed that the hyperhomocysteinemic groupBax protein expression of immune response points compared with the controlgroup was significantly lower, the difference was statistically significant (P <0.05),hyperhomocysteinemic group of Bcl-2protein expression in the immune responseintegral than the control group was significantly l difference was statisticallysignificant (P <0.05); rosuvastatin in the intervention group Bax expression in theimmune response points significantly higher than the Hhcy group, a statisticallysignificant difference (P <0.05); Rosuvastatin intervention group Bcl-2proteinexpression of immune response points compared with the control group wassignificantly lower, the difference was statistically significant (P <0.05), real-timequantitative PCR, mRNA Table Rosuvastatin in the intervention group comparedwith Hhcy Bax2-△△Ct was significantly increased, Bcl-22-△△Ctsignificantly reduced, the difference was statistically significant (P <0.05).Conclusion Rosuvastatin on Hhcy rat aorta of Bcl-2/Bax gene expression byaffecting the thereby improving the process of Hhcy Wistar rat aorticatherosclerosis.Conclusions Rosuvastatin can delay Hhcy rat aortic atherosclerosishardening process through decreasing plasma Hcy concentration and influencingaortic tissue Bcl-2and Bax gene expressions.
