Objective:Based on the compound Duzhenggan Jiegu ointment in treating fracture of clinical application and related theoretical research, through the external bone paste observation of X-ray changes in the healing process of bone tissue and BMP-9, VEGF m RNA and Smad-6 m RNA expression changes of rabbits with fracture, bone paste mechanism described promoting fracture healing. Perfect theory basis of Jiegu ointment bone healing, provide theory support for the development of new drug development of tujia.Methods:⑴The compilation of relevant literature, summarize, summarize bone fracture treatment cream applied to the theoretical basis.⑵The experiment of promote fracture healing: After the establishment of rabbit fracture model, the use of digital experimental animals were randomly divided into two groups: the experimental group and the control groups. The experimental group in their bone lesion topical cream and fixed with plaster; control group did not use drugs, were only plaster fixation. In the third, sixth, tenth weekend as required in accordance with the principles of traditional Chinese medicine treatment of fractures of three points, respectively, X-ray detector, X-ray imaging changes observed fractures in different periods, and then the animals were slaughtered the affected bone specimens using real-time quantitative PCR method(real-time Q-PCR) detection of callus at the BMP-9, VEGF and Smad-6 m RNA expression.⑶ The collected data were statistically analyzed: All data for the use of( SX ±) represents the difference between the groups T test, using SPSS 17.0 statistical software for data processing.Results:⑴Jiegu ointment applied to the whole process of fracture healing. In the experimental fracture healing the whole period, the experimental group was significantly better in general than the control group.⑵In the third,sixth,tenth weekend,the experimental group and the control group had significantly different(P <0.05) in the size of callus and healing of the fracture line. X-ray findings of the experimental group was significantly better than the control group. ⑶In the third,sixth,tenth weekend, In the experimental group compared with the control group, the experimental group BMP-9, VEGF m RNA expression was significantly higher(P <0.05).⑷ In the third,sixth,tenth weekend,In the experimental group compared with the control group, the experimental group Smad-6 m RNA expression was significantly lower than the control group(P <0.05).Conclusions:compound duzhenggan Jiegu ointment in fracture healing process can increase the growth rate of the fracture site callus promote fracture fragments healing; can significantly promote the affected bone tissue BMP-9, VEGF gene expression; significantly inhibited lesion of bone tissue expression of Smad-6 gene, so as to achieve the purpose of promoting fracture healing.
