Objective: To observe the effect of hyperuricemia(HUA) on cognitive function in adult rats and exploring the mechanism of hyperuricemia impact on cognitive function. Methods: Firstly, choose 75 healthy adult male SD rats at the of 8 week whose weight is about 200 ± 20 g; then they were randomly divided into three groups: group Blank(n=25), group Distilled water(n=25) and group Hyperuricemia(n=25). Secondly, adopt lavage methods using yeast extract and ethambutol(uric acid enzyme inhibitors) to establish hyperuricemic model in the group Hyperuricemia. No treatment was done in group Blank; group Distilled water was lavaged with the same amount of water in group Hyperuricemia. Thirdly, choose a fixed time every day to lavage; took the eye blood to measure level of blood uric acid at a fixed time every weekend. At the 7th weekend, morris water maze test was used to measure the learning and memory ability(escaping latency time and passing platform times)(17/group), then randomly choose rats 9/group, separating the hippocampus to homogenate, we took the clear liquid to test the content of MDA, GSH-Px, ASAFR, SOD et al.oxidative stress indicators. Then the left 8/group were dealt with HE staining, hippocampus area neuron cell morphological structure was also observed under the light microscopy. At the same time, the situation of cone-shaped cells apoptosis hippocampus area should be seen through TUNEL as well as cone-shaped cells(AI %). Results: 1.Hyperuricemic model: uric acid level of group Hyperuricemia has reached 285μmol/L at the 3th weekend, and it maintained a high level later, shows a successful building of model. 2. Morris water maze test: 1) as the training time becoming longer, the average escaped latency time to search platform of each group shortened; at the same period,compared with group Blank and group Distilled water, the average escaped latency time to search platform of group Hyperuricemia shows no obviously shortened(P>0.05). 2) compared with group Blank and group Distilled water,passing platform times of group Hyperuricemia shows obviously increased, but there is no statistically significant difference among them(P>0.05). 3. Oxidative stress indicators: compared with Blank and Distilled water group, the MDA of group Hyperuricemia is significantly decreased(P<0.05); GSH-Px,ASAFR,SOD of group Hyperuricemia are significantly increase-d(P<0.05);. 4. Pathology change: hippocampus neurons form and size between groups are basically normal under the HE staining light microscopy. They present round or oval, closely packed, and there is no obviously decrease of cone cell number. Nuclei are pale blue and staining color is well-distributed. What’s more, nucleolus is clearly visible and nuclear membrane is complete. There is no cell edema, no obvious nuclear pyknosis and karyorrhex diseased cells etc. It shows no statistically significant difference among groups. The result of TUNEL shows, there is no obverse apoptosis of hippocampus cone cells, compared with group Blank and group Distilled water, AI% of group Hyperuricemia also has noobversely decreased(P>0.05). Conclusion: 1. Through adopting lavage methods using yeast extract and ethambutol(uric acid enzyme inhibitors) to establish hyperuricemic model, which is very similar to the form and the pathophysiological process of human beings with simple and feasible operation, it is also a relatively and ideal animal model for us to research of hyperuricemia and related diseases; 2. We have not seen obviously effect in ability of study and memory of hyperuricemia for 5 weeks. 3. Suffering hyperuricemia for 5 weeks can effectively reduce the harmful oxidative stress free radicals and increase free radical clearance as well as reduce damage from oxidative stress reaction; 4. We have not seen obviously change in hippocampus neuron cell of hyperuricemia for 5 weeks.
