Background: The prevalence of hyperuricemia and gout is increasing year by year,becoming one of the chronic diseases that seriously endangers human health.As a product of purine nucleotide,uric acid can break the oxidative and antioxidant balance of the body in a variety of ways,so that human is in a state of oxidative stress,which can lead to various related diseases.Melatonin secreted by the pineal gland not only promotes sleep,but also possesses potent anti-oxidative and anti-inflammatory activities.In patients with chronic renal failure,diabetes mellitus and hemodialysis patients,the MT level was reduced.However,there is little research on the level of MT and the relationship between MT level and other oxidative stress indexes in HUA patients.Objectives: In this study,the relationship between melatonin and oxidative stress in patients with hyperuricemia was investigated by detecting melatonin and oxidative stress levels in patients with hyperuricemia.Methods: 181 patients were obtained from Affiliated Hospital of North Sichuan Medical College from April 2017 to November 2017,incuding 40 patients with simple hyperuricemia,40 cases of hyperuricemia combined with hyperlipidemia,31 cases of hyperuricemia combined with diabetes mellitus and 30 cases of hyperuricemia combined with hypertension were hyperuricemia gorup,40 healthy subjects in the same period as normal control group.The levels of MT were detected by ELISA.The levels of MDA,NO,SOD,and GSH-Px were determined by colorimetric assays.Automatic biochemical analyzer to measure liver and kidney function.The levels of melatonin and oxidative stress between the groups were compared,and the correlation between them was analyzed.Results: 1.General clinical data: Systolic and diastolic blood pressure in hyperuricemia with hypertension group were significantly higher than those in the other four groups(P<0.05),HDL-C levels are lower than those in the other four groups;The levels of serum creatinine,urea,AST,ALT,GGT,LDL-C,TG in simple hyperuricemia group,hyperuric acid combined hyperlipidemia group,hyperuricemia combined diabetes group and hyperuric acid combined hypertension group were significantly higher than those in normal group(P<0.05).TC levels in hyperuricemia combined with hyperlipidemia group were higher than those in the other four groups(P<0.05).There was no significant difference in age and BMI between 5 groups(P<0.05).2.Melatonin and oxidative stress Index level: The levels of MT in simple hyperuricemia group,hyperuric acid combined with hyperlipidemia group,hyperuric acid combined with diabetes group and hyperuricemia combined hypertension group were significantly lower than those in normal group(P<0.05),but there was no significant difference among the other four groups(P>0.05).Simple hyperuricemia,hyperuricemia combined with hyperlipidemia,hyperuricemia combined with diabetes mellitus,hyperuricemia combined with hypertension were compared with normal group respectively,MDA level was significantly increased;SOD,GSH-Px levels decreased significantly(P<0.05),However,there was no statistical difference between MDA,SOD and GSH-Px between these four groups(P<0.05).There was a decrease in NO level in the group with hyperuricemia,hyperuricemia combined with hyperlipidemia and hyperuricemia combined with hypertension compared with normal group and hyperuricemia combined with diabetes mellitus(P<0.05).3.Correlation between melatonin and related oxidative stress indexes in hyperuricemia gourp: There was a positive correlation between serum melatonin and GSH-Px(t=5.72,P<0.05),but no significant correlation between melatonin and MDA,SOD,NO(P>0.05).4.Correlation between MT,oxidative stress Index and Clinical data: There was a negative correlation between MT and systolic blood pressure,LDL-C(t=-4.649,P<0.05;t=-2.165,P<0.05).There was no significant correlation with age BMI,diastolic blood pressure,ALT,AST,GGT,serum creatinine,urea,TG,TC,HDL-C.Conclusion: 1.Simple hyperuricemia,hyperuricemia combined with hyperlipidemia,hyperuricemia combined with diabetes mellitus,hyperuricemia combined with hypertension and were in oxidative stress state.2.The levels of MT in simple hyperuricemia,hyperuricemia combined with hyperlipidemia,hyperuricemia combined with diabetes mellitus,hyperuricemia combined with hypertension patients were decreased.3.There was a positive correlation between serum melatonin and GSH-Px in hyperuricemia gourp.
