Objective: In order to make secondary development of Baoguang Fengshi Ye on the market, animal tests in inflammatory, pain and knee osteoarthritis models were carried out respectively in the research to study the effects and mechanism of Baoguang Fengshi Ye. Methods:1.The anti-inflammatory effect of Baoguang Fengshi Ye was analyzed by using the methods as follows: mice ear swelling with xylene, rats paw swelling with egg white, tampon granuloma for mice. 2.The abirritation effect of Baoguang Fengshi was analyzed by using the methods bellow: acetic acid to lead to mice writhing and hot plate. 3.Effects of Baoguang Fengshi Ye on animal models of knee osteoarthritis: ①The rabbit model of KOA was established with anterior cruciate ligament transection. The general morphological changes of synovium, synovial fluid and articular cartilage were observed by nacked eyes. The change of synovium’s pathological changes after HE staining was observed by light microscope. Elisa methods were used to detect levels of SOD and MDA in plasm and MMP-1 in knee joint synovial fluid. ②The rat model of KOA was established with injecting papain into the joint cavity. The general morphological changes of synovium,synovial fluid and articular cartilage were observed by by nacked eyes. The change of articular cartilage’s pathological changes after HE and Toluidine blue staining and synovium’s pathological changes after HE staining was observed by light microscope. Elisa methods were used to detect levels of TNF-α, IL-1β, MMP-1 and MMP-3 in knee joint synovial fluid. Results: 1. Compared with distilled water model group, 0.35,0.70 g native drug/ kg groups of Baoguang Fengshi Ye could has a significant inhaibition on mice ear swelling with xylene; 0.14,0.28,0.56 g native drug/kg groups of Baoguang Fengshi Ye could has a significant inhaibition on rats paw swelling with egg white after 1 hour, 0.28 g,0.56 g native drug/kg groups of Baoguang Fengshi Ye after 2 hour and 3 hour,0.28 g native drug/kg group of Baoguang Fengshi after 4 hour; 0.35,0.70 g native drug/kg groups of Baoguang Fengshi Ye could has a significant inhaibition on tampon granuloma for mice. 2. Compared with distilled water model group,0.17, 0.35,0.70 g native drug/kg groups of Baoguang Fengshi Ye could has a significant inhaibition on acetic acid to lead to mice writhing; 0.35 g native drug/kg group of Baoguang Fengshi Ye could has a signifant inhaibition on hot plate 60 minutes after given Baoguang Fengshi Ye. 3.①In the test of the rabbit model of KOA established with anterior cruciate ligament transaction,compared with distilled water model group: the general morphological changes of synovium,synovial fluid and articular cartilage of treating groups were better and the change of synovium’s pathological changes of treating groups was lesser in different degree. The level of MMP-1 in knee joint synovial fluid of 0.07,0.28 g native drug/kg groups was lower.②In the test of the rat model of KOA with injecting papain into the joint cavity, compared with distilled water model group: the general morphological changes of synovium, synovial fluid and articular cartilage of treating groups were better and the change of articular cartilage and synovium’s pathological changes of treating groups was lesser in different degree.The levels of TNF-α in knee joint synovial fluid of 0.14,0.28,0.56 g native drug/kg groups of Baoguang Fengshi Ye were lower,the levels of IL-1β and MMP-3 of 0.28, 0.56 g native drug/kg groups were lower and the levels of MMP-1 of 0.14,0.28 g native drug/kg groups were lower. Conclusion:Baoguang Fengshi Ye has certain anti-inflammatory effect and abirritation effect. It could improve the pathological changes of rabbit knee osteoarthritis induced by anterior cruciate ligament transaction and rat knee osteoarthritis induced by papain in different degree and reduce the articular cartilage damage and synovium inflamemation.The possible mechanism was reducing the levels of TNF-α, IL-1β, MMP-1 and MMP-3 in knee joint synovial fluid to prevent and treat knee osteoarthritis.
