Objective: Isoniazid is now the most common and important anti-tuberculosis drug. It can kill mycobacterium tuberculosis of cells inside and outside. Would it have influence on the immune function of removing mycobacterium tuberculosis? At present, the problem is not clear. So we design the experiment to study the influence of isoniazid on peripheral blood T lymphocyte(CD3+ T cell, CD4+ T cell, CD8+ T cell), NK cell(CD3-CD161+ cell) and macrophage function of the rat. Methods: 1.clean grade, two months(weight:180±30g), 60 SD male rats were randomly divided into three groups and respectively named fed one month, fed three months, fed three months and fed free one month. Each group was randomly divided into test group and control group. 2.The SD rats grew at the same conditions. The test groups were fed on isoniazid and the control groups were fed on 0.9% NS. 3. The percent of CD3+ T cell, CD4+ T cell, CD8+ T cell and NK cell of rats’ peripheral blood were detected by flowcytometry(FCM), and the clearance index and phagocytic index of macrophage were detected by carbon clearance method.4.The data of CD3+ T cell, CD4+ T cell, CD8+ T cell, NK cell, clearance index and phagocytic index of three control groups at different times were analysed by multiple independent sample each comparison LSD test. The data of every index of test group and control group at the same time wereanalysed by two independent sample t test and tˊ test(heterogeneity of variance).The results were shown in xˉ ± s and P<0.05 was statistically significant. Results: 1.There were not statistically significant differences in CD3+ T cell, CD4+ T cell, CD8+ T cell and CD4+/CD8+ in control group at different times(P>0.05). The statistically significant differences were found in NK cell, clearance index and phagocytic index in control group at different times(P<0.05).The NK cell of control group at fed free one month was higher than that of control group at fed one month(P<0.05). The clearance index and phagocytic index of control group at fed free one month were higher than that of control group at fed three months(P<0.05). The clearance index and phagocytic index of control group at fed three months were higher than that of control group at fed one month(P<0.05). 2.①The CD3+ T cell, clearance index and phagocytic index of test group were higher than that of control group at fed one month(P<0.05). The statistically significant differences were not found in CD3+ T cell, clearance index and phagocytic index of test group and control group at fed three months and fed free one month(P>0.05).②The CD4+ T cell and CD4+ /CD8+ of test group were higher than that of control group at fed one month and three months(P<0.05). The statistically significant differences were not found in CD4+ T cell and CD4+/CD8+ of test group and control group at fed free one month(P>0.05). ③The statistically significant differences were not found in CD8+ T cell and NK cell of test group andcontrol group at the same time(P>0.05). Conclusion: 1. Along with the age increasing, the rats’ NK cell in peripheral blood, carbon clearance ability and phagocytic ability of macrophage might increase. 2. Isoniazid might increase the rats’ CD3+ T cell, CD4+ T cell and CD4+/CD8+ in peripheral blood. Isoniazid might enhance carbon clearance ability and phagocytic ability of rats’ macrophages. Isoniazid had no obvious effects on the rats’ CD8+ T cell and NK cell. 3. Isoniazid not only can kill mycobacterium tuberculosis, but also has positive immune function.
