Objective: To study the influence of isoniazid on peripheralblood cytokines (IL-6, IL-12, IFN-γ) and macrophage function of the rat, and todiscuss the possible influencing mechanism. Methods:1.60clean grade SDrats, male,2months (weight:150±30g), were randomly divided into3groupsand respectively named fed the drug one month, fed the drug three months, fedthe drug three months and one month after drug withdrawal. Each group wasrandomly divided into test group and control group.2. The SD rats were fed atthe same standard conditions. The test groups were fed on isoniazid and thecontrol groups were fed on normal saline.3. The contents of rats’ peripheralblood cytokines IL-6, IL-12, IFN-γ were detected by ELISA and the contents ofrats’ peripheral blood lysozyme were detected by AGAR plate method and theactivities of acid phosphatase in rats’ peritoneal fluid macrophages weredetected by lead nitrate method (Comori method).4. The data of IL-6, IL-12,IFN-γ and lysozyme of test group and control group at the same time wereanalysed by two independent sample t test and tˊtest (heterogeneity ofvariance). The data of three control groups at different times were analysed byq test and results were shown inˉx±s and P<0.05was statistically significant.The data of acid phosphatase of test group and control group at the same timewere analysed by rank sum test and the comparison between three controlgroups at different times were analysed by Kruskal-Wallis H test. It could befurther compared the two which was analysed by Nemenyi test (if there wasstatistically significant) and results were shown in mean rank and P<0.05wasstatistically significant. Results:1. The statistically significant differences werenot found in IL-12, IFN-γ and lysozyme in test group and control group at thesame time (P>0.05).2. There was statistically significant differences in acid phosphatase in test group and control group of fed the drug one month (P<0.05).The test group was higher than that in control group. The statisticallysignificant differences were not detected in others groups (P>0.05).3. Thestatistically significant differences were found in IL-12and lysozyme in controlgroup at different times (P<0.05). We didn’t find the statistically significantdifferences in IFN-γ and acid phosphatase in control group at different times(P>0.05).4. The IL-6didn’t be detected by this method. It was probably due tothe content of IL-6in the experimental rats was lower than the detection limitof the ELISA kit. Conclusion:1. With the rats’ growth, the rats’ serum contentsof IL-12and lysozyme reduced.2. Isoniazid might increase the rats’ serumIL-12and the levels of serum IL-12might restore after isoniazid withdrawal.Isoniazid had no obvious effects on the rats’ serum IFN-γ and lysozymecontents. Isoniazid might transiently enhance the activities of acid phosphataseand couldn’t enhance the activities of acid phosphatase persistently.3. Isoniazidhad relatively small effect on the immune function. We didn’t detect theimmune suppression of isoniazid.
