Objective To study the regulation effect and mechanism of action of Fuzheng granules on immune function and provide the theoretical basis for Clinical treatment on RRTI,by using the method of animal experimental research, and changing the content of arterial blood lymphocyte subsets in immunocompromised rats.Methods Choosing50male SD rats weighting about200g,and dividing them into normal group、model control group、low dose group of Fuzheng granules、medium dose group of Fuzheng granules、high dose group of Fuzheng granules, the latter four were collectively referred to as Low immune function group. The normal group in experiment were orally given2ml distilled water once a day from the fist day to the14th day,and were given physiological saline from the fist day to the7th days,by intraperitoneal injection, dose calculated according to0.2ml/100g,twice the first dose.The low immune function groups were given cyclophosphamide(CTX) with the specifications of20mg/ml from the first day to the7th day,dose calculated according to0.2ml/100g,twice the first dose to make the Model of low immune function rats. Drug given from the first days to the14th day as follows:model control group orally given2ml distilled water once a day;Fuzheng granules low dose group:0.25g/2ml once a day;Fuzheng granules medium dose group:0.5g/2ml once a day;Fuzheng granules high dose group:1g/2ml once a day.After the14th day, all rats were fasted for24hours, taking blood from the abdominal aortic, and detecting the content (%) of CD4, CD8, CD45R, CD161in the arterial blood.by using flow cytometry.Results①Compared with the normal group:The content(%) of CD4, CD8, CD45R, CD161in arterial blood of immunosuppressive group significantly decreased, the difference is statistically significant (P<0.01).②Sompared with the model control group:the content(%) of CD4,CD8,CD45R,CD161in Fuzheng granule low dose group increased,but the differences were not significant (P>0.05);The content(%) ofCD4,CD8,CD45R,CD161in arterial blood of Fuzheng granules in medium dose group increased,there was a significant difference between two groups (P<0.01); The content(%) of CD4,CD8,CD45R in arterial blood of the high dose group of Fuzheng granules increased significantly,and the differences were statistically significant (P <0.01),but the difference of the content(%) ofCD161between the two group was not significant (P>0.05)③Comparison during Fuzheng granules treatment groups:The content(%) of CD4,CD8,CD45R,CD161in arterial blood of Fuzheng granule medium dose group increased statistically (P<0.01),compared to the low and high dose groups;the content(%) of CD4,CD8,CD45R in arterial blood of Fuzheng granules in high dose group increased obviously to the low dose group,these differences were statistical significance (P<0.01),but the difference between the content(%) of CD161was not significant (P>0.05);Compared with the normal group,the contents(%) of CD4,CD8,CD45R,CD161in arterial blood of Fuzheng granules treatment groups were lower,and the middle dose group was the most close to the normal group,but still lower,and the differences were statistically significant (P<0.01)Conclusion①Cyclophosphamide can reduce rats humoral,Immune and cell immunity function.②Fuzheng granules can improve the content of NK cells in arterial blood of Low immune function rats, to enhance innate immunity level,so as to improve the immune function of rats.③Fuzheng granules can improve the content of T lymphocyte subsets in arterial blood of Low immune function rats, to regulate the cellular immunity function, so as to improve the immune function of rats.④Fuzheng granules can improve the Content of B lymphocytes in arterial blood of Low immune function rats, to enhance the level of humoral immunity in rats, so as to improve the immune function of rats.
