Objective: This study through observing rat intestinal Ig A in 5 /6 nephrectomy with chronic renal failure(CRF),and the impact on the intestinal Ig A after administered Turbidity-reducing Granule, so as to explore the improved mechanism. of Turbidity-reducing Granule on the intestinal immune function of rats with chronic renal failure(CRF)Methods: 120 healthy male Wistar rats were randomly selected and divided into sham operation group and operation group, 5/6 nephrectomy to establish the rat model with chronic renal failure. Rats in operation group were randomly divided into 5 groups for administration. high-dose treatment group(HD); dose treatment group(MD); low-dose treatment group(LD); Benazepril control group(B); model group(M). Animals were administered Turbidity-reducing Granule or saline for 60 consecutive days as follows: the low-dose group to Jiangzhuo particles(LD); 4.05 g / kg?d; dose group(MD): to 8.1g/kg?d; the high dose group(HD):16.2g/kg?d; Benazepril group: give 2m L/d(1.2mg / d) Benazepril suspension fed; model control group and the sham group herein are saline 2m L / d orally. The rats were observed before and after treatment of signs and general condition, serum BUN, Scr changes, and pathological changes were observed under microscope. ELISA method were used to detect serum and intestinal SIg A.ELISA method were used to detect serum and intestinal SIg A;Rt-pcr detection of intestinal m RNA expression; Western Blot detection Ig A protein expression in the gut.Results:Turbidity-reducing Granule can reduce the level of serum BUN, Scr in rats with CRF, there were statistically significant differences of Benazepril group and high, medium, low dose of Turbidity-reducing Granule group compared with model group(P <0.05), while there were no significant differences between high, medium, low dosage of Turbidity-reducing Granule group compared with Benazepril group(P> 0.05). there were no statistically significant differences in serum SIg A before and after treatment, but intestinal SIg A down regulated in high, medium, low dose of Turbidity-reducing Granule group and model group compared with sham operation group(P> 0.05). Compared with the model group, Benazepril group, and Turbidity-reducing Granule low, middle and high dose group, intestinal Ig A increased significantly(P<0.05), while intestinal SIg A in Turbidity-reducing Granule middle and high dose group was significantly increased compared with Benazepril group.Intestinal Ig Am RNA expression results show that the shell the captopril group, the treatment group compared with model group, Mr The captopril group, low material particles, intestinal Ig Am RNA expression, high dose group was obviously increased(P < 0.05); Compared with Mr The captopril group material particles of high, middle dose group of intestinal Ig Am RNA significantly increased(P < 0.05). Intestinal Ig A protein expression results are basically identical with m RNA expression results: bei the captopril group, the treatment group compared with model group, Mr The captopril group, material particles of low, medium and high dose group of Ig A protein expression significantly increased(P < 0.05); Compared with Mr The captopril group material particles of high, middle dose group of intestinal Ig A protein expression was also significantly higher(P < 0.05).Conclusion: Turbidity-reducing Granuie can reduce the level of BUN Scr of CRF rats, incress CRF inhibit rats intestinal SIg A, especially for the the group of high, medium dose group, the efficacy is better than Benazepril group.
