Study On Anthocyanins Of Four Rabbiteye Blueberries

The thesis is composed of three chapters. The first chapter studied on the content of the active ingredientand extraction enrichment technology of four rabbiteye blueberry(Gardenblue, Powderblue, Brightwell, Baldwin) from Vaccinium and Semen Trigonellae. The second chapter studied on the qualityof four rabbiteye blueberry. The last chapter summarized progress of anthocyanins structures, separation purification and bioactivities of the last five years.The first chapter, HPLC was used to detectived the cholorogenic acid content and the total anthocyanin concent. Ultrasonic assisted extraction method was used to extract anthocyanins, the total anthocyanin contentl as anindicator, different ethanol concentration, p H, ratio of material to solvent, temperature, extraction time, extraction times were compared on the effect of the total anthocyanin contentl. Five kinds of common materials from laboratory(AB-8, D-101, Rp-18, Sephadex LH-20, silical gel) were compared on the effect of the enrichment to anthocyanin. Experimental results show that, gardenblue express the highest cholorogenic acid content was 0.88%, the other three were about 0.2%. The total anthocyanin concent of four rabbiteye blueberry were between 1% and 4%, the fresh and freeze-dried fruit of gardenblue had the hightest total anthocyanin concent were about 3.02% and 3.18%. The optimum extraction process were as follows p H=2, alcohol contraction 60%, solid-liquid ratiol 1:25, extraction temperature 50℃, extraction time 40 min, extraction times 3, the maximum extraction rate was 0.66%. AB-8 chould used to enrichment anthocyanin and desugared, Rp-18 and sephadex LH-20 chould used to purification anthocyanin. In conclusion,used ethanol ultrasonic assisted extraction method to extracted anthocyanin have the characteristics of simple and efficiently, frist used AB-8 than Rp-18 or sephadex LH-20 to enrichment and purification anthocyanin, so the total anthocyanin concent and yield have risen greatly.The second chapter includes, the anthocyanins of fresh and freeze-dried fruit for the four cultivar rabbiteye blueberry, the ABTS, DPPH, OH· radical-scavenging activity to study the antioxidant activity of four fresh cultivar rabbiteye blueberry, total anthocyanin content of the dried fruit for the four cultivar rabbiteye blueberry before and after the treatment of cold stored and room temperature. Experimental results show that, the kinds of anthocyanins of four cultivar rabbiteye blueberry were more than 13, the kinds of anthocyanins of gardenblue and powderblue were abundant, and all kinds of anthocyanins show blance fractions, the kinds of anthocyanins of brightwell and baldwin were relatively less, and the main anthocyanins represent more than half of total anthocyanins. The gardenblue showed the best antioxidant activity compared with the other three, concentration of 8 mg/ml its antioxidant activity was equal to Vc(8 mg/ml) of antioxidant activity. The anthocyanin under room temperature was instability. In conclusion,the concent of anthocyanin of the four cultivar rabbiteye blueberry were very hight, and the kinds of anthocyanins were abundant, the anthocyanin of the four cultivar rabbiteye express very good antioxidant activity, but the anthocyanin was instability, so the blueberry and the concrete containing anthocyanins had better keep refrigerated and protect from light.The third chapter summarized progress of anthocyanins structures, separation purification and bioactivities of the last five years.
