The Study Of Chaihu Jia Longgu Muli Decotion On Treating Insomnia

Objective:This paper collected material at all times and in all over the world, synthesized various theories,based on the "shaoyang for the pivot" theory and experiment to explore the mechanism of Chaihu Jia Longgu Muli Decotion in the treatment of insomnia,aimed at providing the exact pharmacological mechanism of classical prescriptions for treatment of insomnia.Methods:In the section of theoretical study: Frist,throught combing the ancient literature,summarized the pathogenesis of insomnia is that Yang cannot come into Yin, Yin and Yang never meet. Second,based on the "Shaoyang for the pivot" theory, from the physiological characteristics and pathological manifestations of Shaoyang, demonstrated that Shaoyang is the pivot of three Yang,the pivot of Yin and Yang,the pivot of the movement of Qi, also is the key point of the movement of Qi of Yin and Yang(include ascending-descending and coming in-going out).It is close to sleep. Dysfunction of Shao’ Yang, abnormal movement of Qi, and the non-intersect between Yin and Yang,are the main pathogenesis of insomnia.The pathological changes is that the heart cannot store the spirit, and liver cannot keep the soul. Third,from the aspects of therapeutic methods,demonstrated that Chaihu Jia Longgu Muli Decotion has the function of measuring the shaoyang pivot, calming and placating the soul.In the section of experimental study, maked p-chlorophenylalanine(PCPA) induced experimental insomnia model, and then divided the animals into three groups:the normal control group, model group,Chaihu Jia Longgu Muli Decotion group(TCM group).Observing the insomnia model rats’behavior and detecting the amount of IFN-γ、IL-1β、IL-4、IL-10 and IFN-y/IL-4. Inorder to study the relationship between insomnia and balance of the Thl/Th2,and the effect of balance of the Thl/Th2 after treatment of Chaihu Jia Longgu Muli Decotion.Resule: 1.Behavioral performance: No significant change in normal control group;the sleep-wake rhythm disappeared in model and TCM group at 30h after the first intraperitoneal injection of PCPA suspension, in TCM group rats, insomnia symptoms were gradually restored after lavaging of Chaihu Jia Longgu Muli Decotion compared to model group. 2. Weight change:before administered, the weight of model group are equivalent to TCM group’s, but both lower than the control group’s(p<0.01);before taking blood, TCM group’s than model group’s increased(p<0.05). 3. Detection: After modled,compared with the control group, in model group’s the content of IFN-y and IL-1β decrease, IL-4 and IL-10 increase, IFN-γ/IL-4 decrease (P＜0.05); the balance of Thl/Th2 move towards Th2; after treated, compared with the model group, in TCM group’s the content of IFN-γ and IL-1β increase, IL-4 and IL-10 decrease, IFN-γ/IL-4 increase (P<0.05); the balance move towards Thl.Conclusion: 1. Chaihu Jia Longgu Muli Decotion is the first choice of prescription on treating insomnia caused by the dysfunction of Shaoyang pivot. 2. Chaihu Jia Longgu Muli Decotion can improve the symptoms of insomnia rats, speculating the mechanisms on treating insomnia may affect the immune system, so that the balance of Thl/Th2 move towards Thl to promote of sleep.
