Objective:To investigate the effect of Liuweidihuang pill on JAK/STAT signaling pathway gene expression in postmenopausal osteoporosis with the kidney yin deficiency, and to explore the molecular mechanism of Liuweidihuang pill in the treatment of postmenopausal osteoporosis with the kidney yin deficiency.Methods:According to the TCM syndrome,6 women with the kidney yin deficiency and postmenopausal osteoporosis were randomly selected. Another 3 healthy postmenopausal women were selected as control group. Patients with the kidney yin deficiency were treated with Liuweidihuang pill for 3 months. The lever of gene expression in JAK/STAT signaling pathway was detected using special chip. The exist of significant difference between the groups was determined by △△Ct method.Results:1. Compared with those in control group, the expression of 4 differential genes was down-regulated in the kidney yin deficiency group before the treatment, including PRLR, JUN, JUNB, and INSR.2. After the treatment,25 differentially expressed genes were identified in the kidney yin deficiency group. Among these 25 genes,11 genes were up-regulated, and 14 genes were down-regulated.3. The expression of JUN and JUNB was up-regulated significantly.(1) According to the analysis of the biological function of the differentially expressed genes, the immune response genes included PRL, PRLR, OSM, ISG15, and IL4R.(2) The cell growth and/or maintenance genes included F2 and SOCS3.(3) The transcription factor or regulator genes that interact with STAT proteins included JUN, JUNB, SMAD3, SMAD5, SMAD4, SP1, and YY1.(4) The negative regulators of the JAK/STAT pathway included PIAS1 and SOCS4.(5) The SH3/SH2 adaptor genes included SRC and STAM. Conclusions:The mechanism of Liuweidihuang pill in the treatment of postmenopausal osteoporosis with the kidney yin deficiency may be related to the regulation of the genes in JAK/STAT signaling pathway. These genes are involved in immunity function, cell proliferation differentiation, and the growth of osteogenesis.
