Objective:To explore quality research of NZ compound extracts and preparation processoptimization research, to guide the development of NZ compound and similar providereference for the study of TCM syndromes.Method:①Study the pharmacodynamics of microbial and chemical composition analysis of NZcompound extracts, then combine with the results of the two to determine thecorresponding relation of the index ingredients P with the pharmacodynamics of microbialof the NZ compound extracts.②The preparation process of NZ compound extract and process optimization are studied.The first step: By comparing the different extraction methods of NZ compound extract theinfluence of the preparation. Analysis the index ingredients P by determined correspondingrelationship between the index ingredients P with the pharmacodynamics of microbial ofthe NZ compound extracts, then select NZ extract optimum extraction method. The secondstep: Study solvent, dosage of solvent, crushing, extracting times and extracting time fivesingle factor’s influence on NZ side extract preparation technology, chose to dosage ofsolvent (A)、extracting time (B)、extracting times (C) as the influencing factors, selectedthree factors and three levels of orthogonal test design table L9(33) orthogonal testoptimization process, select the best preparation technologyⅠ.The third step: on the basisof processⅠ,combine with the demand of industrial production materials in energy saving,using the integrated test method, study the dosage of solvent and extracting time at thesame time to get the best preparation process Ⅱ.Magnifying process Ⅱ and processⅠ,analysis the index constituents P and study extracts pharmacodynamics respectively, finallydetermine the NZ compound extracts the best preparation technology. Result:①The index ingredients P chemical composition of NZ compound extracts is relativelystable, and it can can establish ultraviolet analysis method and HPLC analysis method; NZcompound extracts have good antibacterial effect on the deputy hemolytic bacteria anddissolve algae bacteria, and the content of index component P have a positive relationshipwith NZ compound extracts’ bacteriostatic efficacy.②The best preparation process of NZ compound extracts that comes from the result of thepreparation process optimization research is processⅠ: adopt heating reflux extraction;solvent Ⅴ; extracting3times;extracting time for each time45min/45min/15min;extraction menstruum dosage for each time6times/5times/4times.③NZ compound extracts made by the best preparation technology on extraction of NZcompound medicinal herbs have bacteriostatic efficacy: minimal inhibitory concentrations(MIC) to dissolve algae bacteria is7.8mg/mL, the minimal inhibitory concentrations((MIC) of deputy hemolytic bacteria is1.95mg/mL; the content of index component P inNZ compound extract as follows:301.89mg/mL.Conclusion:①The index component P content and therapeutic effects of NZ compound extractsare positively related, and it can provide a feasible evaluation method for the preparation ofNZ compound extracts.②Evaluate NZ compound extracts preparation technology with the positive correlation ofindex component content P and efficacy, optimize processⅡas the best preparation.The extracts has high content of P and good therapeutic effects from technological Ⅱ inNZ medicinal herbs extract.Therefore, use process Ⅱ in NZ compound medicinalmaterials may has the advantages of low cost and small energy consumption in theindustrial production.
