ObjectiveSelect the best anti-infective traditional Chinese medicine prescription QC,for the prevention and treatment of infectious diseases to offer reference to the research and development of new drug of TCM.Methods(1)By Kirby-Bauer and broth microdilution method,to extract the Deputy hemolysis vibrio,Soluble v,Aeromonas hydrophila,E.Coli,Streptococcus pneumoniae,staphylococcus aureus,6 kinds of pathogenic bacteria of bacteriostatic ring size and the minimum bacteriostasis concentration as an index,establish pharmacodynamic evaluation method,select the best of traditional Chinese medicine compound to fight infection.(2)Ultraviolet-visible spectrophotometry is used to establish traditional Chinese medicine prescription QC chemical evaluation method,the compound QC extraction method,solvent,extraction time and extraction times,with the various single factor level.Using the orthogonal design experiment of L9(34)with 4 factors and 3 levels optimized traditional Chinese medicine prescription QC the best extraction technology,combined with the index component transfer rate,the effects of the extract and comprehensive evaluation,the production cost for the preparation of compound extracts from QC process.(3)Thin layer chromatography(TLC)is adopted to establish traditional Chinese medicine prescription QC index component Q qualitative identification method.Establish a high performance liquid chromatography(HPLC)method for testing the content of compound QC index component Q.Result(1)Compound QC,QC2,QC3 bacteriostatic ring of various pathogenic bacteria can be up to 19 mm in diameter,minimum bacteriostatic ring diameter of 8 mm,has good bacteriostatic action.Screening for the best anti-infective is compound QC,the above-mentioned six kinds of pathogenic bacteria of MIC are: 3.13 mg/mL,3.13 mg/mL,3.13 mg/m L,12.5 mg/mL,6.25 mg/mL,6.25 mg/mL.(2)Compound QC extract the maximum absorption wavelength of ultraviolet is 289 nm.Linear investigation,repetitive inspection,stability test,sample recovery rate methodology such as RSD value is less than 2%.The experimental results confirmed the solvent ? the best solvent for extraction process.And to determine the maximum content of prescription in the QC index component Q is 83.79 mg/g.Orthogonal experiment the result is: the medicinal material crushing grinding of 40 mesh,adding solvent ? reflux extracting 3 times,each time 45 min,every time to join the dosage of solvent are all 12 times.The menstruum dosage of modified to add each time reduced to 10 times,eight times,six times.In the comparative experiments,using the orthogonal process and modified process to extract the constituent of compound QC get index Q transfer rate of 92.89%,91.63% respectively.Herbal extracts on Deputy hemolysis vibrio,Soluble v,Aeromonas hydrophila the MIC of bacterium is 0.391 mg/mL,MIC of streptococcus pneumoniae,golden staphylococcus were 0.781 mg/mL,for e.coli MIC is 1.56 mg/mL.(3)With n-butyl alcohol-trichloromethane-methyl alcohol(4:4:3)as agent,reference substance solution point sample is 1 ul,point sample amount of test sample solution for 5 ul,thin layer chromatography(TLC)was adopted to construct compound QC qualitative identification method.In this deployment under the condition of the spots were clear,the Rf value is moderate,the method is simple,sensitive.(4)Chromatographic conditions using the following: the mobile phase: 0.1%phosphoric acid solution and acetonitrile;Chromatographic column: Agilent Eclipse Plus C18(4.6 x 250,5 microns);Column temperature 35?;detection wavelength of 267 nm;Detector: DAD;The analysis of time: 25 min;Speed: 1 mL/min.Improved technology was used to extract,the extract use HPLC to measure index component content is 75.75 mg/g,transfer rate was 90.40%.Results of UV transfer rate was 91.41%.Both the extracts onDeputy hemolysis vibrio,Soluble v,Aeromonas hydrophila the MIC of bacterium is 0.391mg/m L,MIC of streptococcus pneumoniae,golden staphylococcus were 0.781 mg/mL,for e.coli MIC is 1.56 mg/mL.ConclusionThis subject selection of compound QC has good anti-infective effect.Modified process to extract the constituent of the index Q transfer rate is the largest,it can be stable at around 91%.This can provide experiment basis for the development of new anti-infective Chinese medicine in the future.
