Melilotus officinalis (Leguminosae) is an important medicinal plant distributed worldwide. Data from traditional phytotherapy, show that this plant had been used to diminish inflammation and as a treatment for detumescence, diuresis, hemorrhoids and hemorrhoids syndromes.In this paper, we began to study the chemical constituents of the aerial parts of Melilotus officinalis (L.), six compounds were isolated and purified from the alcohol extract of Melilotus officinalis by silica gel column chromatography and re-crystallization. On the basisof various modern spectral analysis (1H-NMR,13C-NMR) and classical chemical methods,structures of six compounds were identified as triacontanol (1), coumarin (2), umbelliferone (3), 6,7-dimethoxycoumarin (4), kaempferol (5) and daidzein (6). Compound (6) daidzein were isolated from this plant for the first time.A reversed-phase high performance liquid chromatography (RP-HPLC) method was developed for the determination of coumarin in Melilotus officinalis. The content of coumarin was analyzed through different batches, different medical parts and different growth periods samples. In addition, we identified the samples by TLC. The content of coumarin was different among the tested samples from the different production areas.The preparation process of the Melilotus officinalis extract was investigated by a systematic study. Furthermore the preparative method of Melilotus officinalis extract was discussed as well. The coumarin assay of Melilotus officinalis extract was established by RP-HPLC method. The results showed that the RP-HPLC method is accurate and reliable.The results will provide pharmaceutical and technological bases for the development of this medicine.
