【Objective】To evaluate the significance of letrozole-induced PCOS rat model anddepression rat model induced by chronic unpredictable mildstress. To find outimpacts of PCOS and depression on both serum sex hormone and Neuro-transmitterTo analyse of the relationship between depression and PCOS, to provide thereferencefor clinical research.【Method】Using the models we bulit in partⅠand partⅡexperiments, the fortyfemale SD rats were divided into four groups: normal control group(n=10),depression group(n=10),PCOS group(n=10) and depressionPCOS group (n=10).Serum T and LH levels were measured by radioimmunoassay. The levels of GnRH,NE,DA and5-HT in hypothalamic were measured by ELISA and HPLC-fluorescence.【Results】1. The letrozole-induced PCOS rat model showed similar clinical andendocrine features with human PCOS.2. The depression rat model induced by CUMS can successfully mimic clinicalsymptoms of depression.3. The difference of rats serum LH and T levels were statistically significant amongthe four groups(P<0.05), and there was a gradual increasing trend. The serum LHand T levels in groups of PCOS and PCOS with depression were higher than those ofnormal control group.The serum LH and T levels were higher in depressionPCOS group than depression group(P<0.05).4. The rats’ GnRH levels among the four groups in the hypothalamus compared withstatistical significance(P<0.05), and there is a gradual increasing trend. PCOS groupand the normal control group, PCOS group and the normal control group, PCOS withdepression group and normal control group,PCOS with depression group and thedepression group all have significant differences.(P<0.05). 5. DA, NE and5-HT levels in hypothalamus were singificantly different among4groups(P<0.05), NE trend to increase, whereas DA and5-HT tend todecreese.(1) Nuerotransmiter DA is singificantly different bewteen PCOS group andthe normal control group(P<0.05).Depression PCOS group was significantly differentwith normal control group(P<0.05).(2) Neurotransmiter NE is significantly differentbetween depression PCOS groups and the normal control group(P<0.05).(3)5-HT level is significantly different among4groups(P<0.05).6. Correlation analysis showed that the level of GnRH in each group had a positivecorrelation with NE(P <0.05, r=0.809).The level of GnRH was negatively correlatedwith DA(P <0.05, r=-0.874)and5-HT(P <0.05, r=-0.821).【Conclusion】1. The PCOS rat model induced by Letrozole is a suitable model forstudying the PCOS neuroendocrine changes.2. The depression rat model which induced by chronic unpredictable mild stress ismore appropriate one to study the neuroendocrine changes of depression patients.3.PCOS and depression are two importance factors that may affect theneuroendocrine system in SD female rats.The abnormal secretion of DA, NE and5-HT may affect the secretion of hypothalamic GnRH and serum LH, T and induceclinical manifestation and psychological and ovarian changes in female rats.The twofactors may exist reciprocal causation relation of mutual promotion. Depression maybe the PCOS occurrence and can cause longterm complications,and PCOS can alsolead to the occurrence of depression and aggravate its development.
