Objective:this topic proposed to observing the effect of circadian rhythm change caused by extending light in artificial light environment on intraocular pressure, expression of melatonin (Mel) both in blood and retina of rabbit, investigating the relationship between intraocular pressure and Mel and intervention function of traditional Chinese medicine guided by the clock adjustment theory of Yin and Yang Methods:healthy New Zealand rabbits1months old, at various stages of the research is divided into control group, model group, Chinese medicine group, both have male and female, are raised in the artificial light environment. Control group:artificial light environment illumination time is set in the surf-18:00, light/dark time each day for12h/12h. Model group, the Chinese medicine group illumination time setting in the surf-24:00, light/dark time every day for18h/6h. Chinese traditional medicine group in daily10:00to fill Yang fang,22:00to fill Yin fang; Experimental specimen collection and data collection time is:the control group and model group in2months and4months of age, Chinese traditional medicine group for4months, specimens at different times of day gathering. Line weight respectively to different groups of animals, the blood and retina specimens, Mel content enzyme-linked immunoassay detection, intraocular pressure detection.Results:ELISA test shows:the control group and model group Mel content in blood and retinal appeared significant difference (p<0.01); Model group in the retina Mel significantly lower than that of control group (p<0.01), Mel expression in the retina Chinese medicine group were higher than in model group (p<0.01), but lower than that of control group (p<0.05), model group and dark environment intraocular pressure waveform is disorder, Chinese traditional medicine group dark environment intraocular pressure waveform is close to the control group but wave than the control group in advance.Conclusion:night and day, Mel content in the blood fluctuation and rabbits diurnal intraocular pressure fluctuation and clock of Yin and Yang theory of TCM the day and the ebb and flow of Yin and Yang in the law of consistency; Extended light source under the condition of same illumination time caused by changes in rhythm can inhibit blood and Mel synthesis in the retina and cause rabbit24-hour intraocular pressure waveform disturbances; Traditional Chinese medicine (TCM) can affect the Mel expression in the retina; Chinese medicine intervention on the day and night environment change YouTu intraocular pressure circadian fluctuations caused by disorder have certain adjustment. Relevant mechanism need further study. |
