Objective:We observed the effects of different light sources on the expression of retinal clock gene Cry1?Cry2 of New Zealand immature rabbits.Based on the theory of yin and yang proposed by Professor Wang Yuliang,we explored the preventive function of traditional Chinese medicine(TCM)"Rizhong Yinyang Fang"intervention proposed by Professor Wang Yuliang on the expression of retinal clock gene Cryl,Cry2.Methods:72 one-month-old New Zealand immature rabbits were randomly divided into two groups with both male and female rabbits in each group.Group 4 weeks and group 12 weeks,respectively.Sunlight group was exposed in natural light while LED group and full spectrum group were exposed in LED and full spectrum light.Based on this,sunlight+TCM group,fluorescent+TCM group and full spectrum+TCM group were added with"Rizhong Yinyang Fang"intragastric administration based on "Theory of yin-yang clock"of Professor Xia Guicheng,respectively.The method of administration was asfollows:The "Yang-fang"was administrated in 10 a.m and the "Yin-fang"was given in 22 p.m.The dosage of young rabbits was converted to the equivalent dose ratio of body surface area according to the body surface area in accordance with the pharmacological experiment,according to the standard of 70kg,A pair of Chinese medicine granules with 300ml water,two times a day oral calculation,adult rabbits(1.5g)each medication 7ml,rabbit 3.5ml gavage needle for each half.The sunlight group,LED group and full spectrum group were not fed with normal saline.Optical parameters were measured.Cryl and Cry2 were extracted from rabbit retina in 4 weeks and 12 weeks.Results:After 4 and 12 weeks,RT-PCR gene:The Cry1 mRNA and Cry2 mRNA levels in the retina of LED group decreased significantly(P<0.01).The content of Cryl mRNA?Cry2 mRNA in rabbit retina of LED group was significantly higher than that of LED group(P<0.01).The traditional Chinese medicine has no effect on the Cry1 mRNA and Cry2 mRNA of the rabbit retina in the natural light group and the full spectrum group(P>0.05)(mRNA).Western blotting showed that the Cryl protein and Cry2 protein levels in the retina of LED group decreased significantly(P<0.01).The content of Cryl protein and Cry2 protein in retina of LED lamp group was significantly higher than that of LED group(P<0.01).The traditional Chinese medicine had no effect on the content of Cryl protein and Cry2 protein in the retina of the natural light group and the full spectrum group(P>0.05).Conclusion:LED lights have a certain impact on the expression of retinal clock gene Cry1?Cry2.The effect of full spectrum lights on the expression of retinal clock gene Cry1?Cry2 is similar to sunlight.The traditional Chinese medicine "Rizhong Yinyang Fan"can intervene the expression of retinal clock gene Cry1?Cry2 when in LED lighting environment.
