ObjectiveMicroRNA (miRNA) is a kind of small molecular endogenous foundin recent years of non coding RNA, through the mRNA3’-UTR and thetarget gene binding play a post transcriptional regulation,with similaroncogenes or tumor suppressor genes. Research shows that miRNAabnormal expression in many malignant tumors, in ovarian cancer tissuemiR-125a expression level increased, the expression level of miR-145decreased, suggesting that miRNA may be associated with ovarian cancer,but the mechanism by which have not reported in ovarian cancer. Thetumor suppressor gene p53as common tumor,its coding p53protein is anegative regulator of cell cycle, suggesting that miR-125a-5p may enhancethe ability of proliferation and invasion of cells by down regulating theexpression of miR-145gene p53, may be to reduce the abilityofproliferation and invasion of cells by up regulating the expression ofgenes p53.In this study, through the establishment of over expressed inovarian cancer cell line SKOV3miR-125a-5p and miR-145,ofmiR-125a-5p and miR-145on cell proliferation, metastasis,invasion andapoptosis effect, provides the basis for further study of miR-125a-5p andmiR-145target genes associated with ovarian cancer. MethodThe lentiviral transfection plasmid containing miR-125a-5p andmiR-145weretransfected into SKOV3cells in two groups by puromycinselection, establishment of stable SKOV3cell line expressing miR-125a-5pand miR-145. By real-time fluorescence quantitative PCR to verify theexpression of two genes in the cells,MTT assay, flow cytometry, Transwellexperiment, scratch test the upregulation of miR-125a-5p and miR-145SKOV3cells in proliferation, apoptosis, invasion and metastasis of thechange, and detected by Western-blot in SKOV3cells p53protein changesexpression of miR-125a-5p and miR-145.Result1.MTT results showed that over expression of SKOV3cells, theproliferation ability of miR-125a-5p significantly increased than the controlgroup, theproliferative capacity of SKOV3over expression of miR-145decreased significantly (P <0.05).2.Detection of flow cytometry showed that over expression ofmiR-125a-5p in SKOV3cell proportion of cells in S phase increased, thepercentage of G0/G1phase cells decreased, the apoptosis rate decreased;the over expression of miR-145in SKOV3cell proportion of cells in Sphase was decreased, the percentage of G0/G1phase decreased, theapoptosis rate increased (p <0.05).3.The scratch test shows that the over expression of miR-125a-5p24hand48h SKOV3cell migration degree are respectively2.37times and4.28times higher than the control, the over expression of miR-125a-5p inexperimental group were higher than those in the control group, the cellmigration ability (P<0.05). 4.transwell experimental results show, over expression of SKOV3cell24h miR-125a-5p than the control group, the number is1.86times higherfor invasion,invasion ability is enhanced; over expression of SKOV3cell24h miR-145than the control group, the number is1.86times higher forinvasion, invasion abilitydecreased significantly (P <0.05).5.Experimental of western-blot results show that the over expressionmiR-125a-5p of SKOV3cells p53protein expression is only64.04%in thecontrol group; the over expression of miR-145in SKOV3cells p53proteinexpression is only64.04%in the control group (P <0.05).ConclusionOverexpression of miR-125a-5p can promote SKOV3cellproliferation, migration and invasion, and inhibition of apoptosis, whichmay reduce the expression of ovarian cancer SKOV3cells in tumorsuppressor gene p53to increase cellmalignant tumor. Overexpression ofmiR-145can restrain SKOV3cell proliferation, migration and invasion,and inhibition of apoptosis, which may promote the expression of ovariancancer SKOV3cells in tumor suppressor gene p53to reduce cellmalignanttumor.
