Objective Though compare mi R-125 a and A20 expression level in peripheral blood mononuclear cells(PBMCs) between patients with ankylosing spondylitis(AS) and healthy controls, and analysis the relationship between mi R-125 a or A20 expression level and AS the clinical features or related cytokines, to find the role of mi R-125 and A20 in the pathogenesis of AS and provide basic data for the diagnosis, treatment and prognosis of AS.Moted With the method of case-control design, from April 2014 to September 2015, we collected 40 AS cases and 40 healthy controls. AS cases from outpatient service of rheumatism and immunology of the first affiliated hospital of anhui medical university, the healthy controls witch frequency matched with AS cases for age and gender were from huaihe river road pedestrian street who donate blood. The patients got 4 or more points of Ankylosing spondylitis disease activity score(BASDAI) were assigned in AS stable group, BASDAI score<4 points were assigned in AS active group. We collected 5 ml venous blood from all cases and controls, separated PBMCs, extracted RNA, and then reverse transcription them to c DNA. Using real-time fluorescence quantitative polymerase chain reaction(RT-q PCR) to detect mi R-125 a, A20 m RNA, IL-12 m RNA and IL-17 m RNA expression levels in PBMCs of AS patients and controls, comparing this expression levels between AS case group and control group, and AS stable groups and the AS active group, and then analyze the correlation between mi R-125 a and A20 m RNA. Using enzyme linked immunosorbent assay(ELISA) test related cytokines IL-1, IL-6, IL-12, IL-17, IFN-β and TNF-αexpression levels in the peripheral blood of AS cases and controls. Analyses the correlation between A20 m RNA、mi R-125 a and the related cytokines. The t test was used to compare between groups, correlation analysis of statistical processing using the Spearman rank correlation.Result In cases group, there were 29 male and 11 female and average age was 28.10±7.86 years old. In control group, there were 28 male and 12 female and average age was 27.32±7.48 years old. Moreover, there was no significant difference between the groups of age and gender. We compared the expression level of mi R-125 a between case group and control group found that no statistically significant difference between the two groups(Z=0.509, P=0.509), and no statistically significant was found difference between AS stabe group and AS active group, too(Z=1.529, P =1.529). In addition, we coulden’t found correlation between mi R-125 a and A20 m RNA(rs=0.030, P=0.804). But mi R-125 a was positively related with IL-6(rs=0.518, P=0.518), IL- 10(rs=0.363, P=0.363), and TNF-α(rs=0.365, P=0.365). Compared with control group, the A20 m RNA expression level of AS case group was increased(Z=3.908, P<0.001), suggesting that exist exist abnormal expression of A20 in PBMCs of AS patients. Between AS stabe group and AS active group there was no Significant differences(Z=0.498, P=0.498). But A20 correlation analysis found that A20 m RNA level negatively correlated with CRP(rs=0.508, P=0.009) and BASDAI(rs=0.352, P=0.352). Expression level of IL- 12 in AS cases group is significantly higher than the controls(Z=5.04, P<0.001), and the expression level of IL-17 in case group is significantly higher than the control group,too(Z = 2.05, P=2.05). In addition, IL-12 positively correlated with IL-17(rs= 0.571, P<0.001).Conclusion We coulden’t observed in abnormal expression of mi R-125 a in PBMCs of AS patients, but A20 abnormal expression in AS patients may play an important role in the pathogenesis of AS.
