BackgroundIncreasing evidence shows that miR-125a-5p could act as a tumor suppressor in several cancers,however,little is known about its mechanism in human esophageal squamous cell carcinoma(ESCC).Objective(1)To Screen abnormal expression of miRNA and functional verification in esophageal squamous cell carcinoma patients.(2)To investigate the potential target genes of miRNA..Methods(1)The expression level of miRNA in ESCC was detected by qRT-PCR.(2)In vivo and in vitro functional experiments were conducted to investigate the effects of miRNA on biological behavior of ESCC.(3)Dual luciferase assay was used to investigate the mechanism of miRNA in ESCC.ResultsWe found that the expression level of miR-125a-5p is significantly reduced in clinical ESCC tissues compared with normal tissues,as well as in the serum of ESCC patients compared with that of healthy volunteers.And it is correlated with T stage and TNM stage.Biological behaviors experiments that in both TE-1 cells and ECA109 cells overexpression of miR-125a-5p inhibits cell proliferation,the G1/S phase transition,and suppresses cell migration and invasion in vitro and in vivo.Moreover,we found that IGSF3,which was reported to associate with cell invasion and metastasis,was a potential target gene of miR-125a-5p by Dual luciferase assays.Overexpression of mi R-125a-5p in both TE-1 cells and ECA109 cells caused an inhibitor of IGSF3 mRNA and protein.Knocking down IGSF3 in mi R-125a-5p over-expressed cells could partially assist the effect of miR-125a-5pon migration and invasion.ConclusionWe are the first to report the interaction between miR-125a-5p and IGSF3 in ESCC and in our study verify new insights into the role of mi R-125a-5p as a biomarker in the diagnosis and treatment of ESCC by targeting IGSF3.
