Objective: To detect the expression of miR-125a-5p in tumor tissues of diffuse large B cell lymphoma,explore the effect of miR-125a-5p expression on the development and prognosis of disease,and the relations with drug resistance tumor protein P-gP expression,thereby,provide experimental data for finding novel therapeutic target and new prognosis related markers.Methods: Use real-time fluorescent quantitative PCR to detect miR-125a-5p mRNA relative expression;immunohistochemical staining detection of P-gP and Ki67 protein expression in tumor tissue.Collection of the clinicopathological data and follow-up were done.Statistical analysis,including the relationships between the expression of miR-125a-5p and P-gP protein expression,proliferation of tumor cells,as well as clinicopathological features and prognosis,were done.Results: mi R-125a-5p expression was significantly higher in DLBCL than the reactive hyperplasia of lymph node(P=0.001);The expression is associated with serum LDH level(P=0.020)and immunophenotypes of tumor cells(P=0.009),whereas it isn’t related to age,gender,primary site,staging,IPI score,all P values were more than 0.05.The positive expression rate of P-gP protein was higher in the group with high expression of miR-125a-5p [29/47(61.7%)] than those with low expression of mi R-125a-5p [17/43(39.5%)],the difference had a statistical significance(P=0.036).51 of 90 cases with DLBCL were followed up.Follow-up rate was 56.7%.Kaplan Meier and log-rank survival analysis showed that there were obviously difference in median survival time and relapse-free survival time between high and low expression of miR-125a-5p groups(34months vs 65 months;36 months vs 60 months,successively)(P=0.048,0.001 successively).No relationship was found between the prognosis and age,gender,serum LDH,IPI score,immunophenotypes,as well as Ki67 expression.COX regression model analysis showed that both ann arbor stage III+IV and miR-125a-5p high expression are independent factors of poor prognosis of DLBCL(P=0.002,P=0.017 successively).Conclusion: The expression of miR-125a-5p in DLBCL is up-regulated,especially in the subtype of non-GCB DLBCL.The expression of miR-125a-5p in DLBCL maybe associated with P-gP protein expression.The high expression of mi R-125a-5p can be considered as an independent poor prognosis related factor of DLBCL.
