ObjectiveDetecting the expression of the Claudin-7protein and the MMP-2protein in Bowen’s disease (BD) and Squamous cell carcinoma (SCC),analyzing the relationship between Claudin-7and MMP-2, to explorethe role of the Claudin-7and the MMP-2protein in the occurrence anddevelopment of infiltrating breast cancer.MethodWe use immunohistochemical SP method to detect the expressionand distribution of Claudin-7and MMP-2protein in20cases ofBowen’s disease,36cases of skin Squamous cell carcinoma(including22cases of high differentiated SCC and14cases ofmoderately differentiated SCC) and10cases of normal skin tissue. Atthe same time, we use Western blot method to detect the expressionlevel of Claudin-7and MMP-2protein in Bowen’s disease, skinSquamous cell carcinoma and normal skin tissue.Results1.The positive rate of Claudin-7protein in normal skin tissue,Bowen’s disease and Squamous cell carcinoma were100%,55.00%,22.22%, respectively. The positive rate of Claudin-7in highdifferentiated SCC and moderately differentiated SCC were27.27%and14.29%. The expression positive rate of Claudin-7protein inBowen’s disease, Squamous cell carcinoma group were significantly lower than normal skin tissues (P<0.05). The expression positive rateof Claudin-7protein in Bowen’s disease group was higher than that ofSquamous cell carcinoma group, and the difference was statisticallysignificant between them (P<0.05). The expression positive rate ofClaudin-7protein in Bowen’s disease, high differentiated SCC andmoderately differentiated SCC is on the decline, and the differencewas statistically significant (P<0.05).2.The positive rate of MMP-2in normal skin, Bowen’s disease andSquamous cell carcinoma were10%,65%and75%, respectively. Thepositive rate of MMP-2in high differentiated SCC and moderatelydifferentiated SCC were72.73%,78.57%. The expression positive rateof MMP-2protein in Bowen’s disease, Squamous cell carcinoma groupwere significantly higher than normal skin tissues (P<0.05). Theexpression positive rate of MMP-2protein in Bowen’s disease groupwas lower than that of Squamous cell carcinoma group, but thedifference was not statistically significant between them (P>0.05).The expression positive rate of MMP-2protein in Bowen’s disease,high differentiated SCC and moderately differentiated SCC was onthe rise, but the difference was statistically significant (P>0.05).3.The expression of Claudin-7and MMP-2protein in Bowen’sdisease and Squamous cell carcinoma showed a negative correlation(r=-0.362,P<0.05).4.Western blot results: we used the Gel-pro analyzer4softwaresystem to analyze the stripe. The results show that the protein level ofClaudin-7protein in normal skin tissue is significantly higher thanBowen’s disease and Squamous cell carcinoma (P<0.05). The proteinlevel of MMP-2protein in normal skin tissue is significantly lower thanBowen’s disease and Squamous cell carcinoma (P<0.05). The proteinlevel of Claudin-7and MMP-2protein in normal skin tissue, Bowen’sdisease, high differentiated SCC and moderately differentiated SCCare respectively in the fall and rise trend, but there was no statistically significant difference (P>0.05).Conclusions1.The expression of Claudin-7in Bowen’s disease and Squamouscell carcinoma is in the fall, and its expression level is positivelycorrelated with tumor differentiation degree, the worse thedifferentiation, lower expression.2.The expression of MMP-2in Bowen’s disease and Squamouscell carcinoma is rising, but its expression level is no positivelycorrelated with tumor differentiation degree.3.The expression of Claudin-7and MMP-2in Bowen’s diseaseand Squamous cell carcinoma showed a negative correlation. Thelower expression of Claudin-7and high expression of MMP-2may berelated to the occurrence and development of Bowen’s disease andskin Squamous cell carcinoma invasion.
