Therapy Of Symptoms And Root Causes For Mice Of Type2Diabetes Induced By High Fat Food

Objective:To investigate the mechanism of "tonifying kidney and strengthening spleen" and" clear heat and dry dampness" treating type2diabetes.Methods:4weeks old male C57BL/6mice were divided into three groups randomly, group negtive control(NC)、group high fat (NF)、group early tonifying kidney and strengthening spleen(EKS). Group NC mice are fed with ordinary food、free of water, while other mice are fed with high fat food. The group high fat mice are divided in group HF、group clear heat and dry dampness (CD) and group late tonifying kidney and strengthening spleen(LKS). The "clear heat and dry dampness" drug we used is berberine, and the "tonifying kidney and strengthening spleen"drug is clinical prescription. The mice were fed with drugs until the20th week when OGTT are taken to test their blood glucose. All the mice are killed at the20th week, and the serological test and the pathological examination are carried out. The mRNA level of the inflammatory factors in liver、fat、muscle tissues are tested by R-T PCR. HE To investigate the Inflammatory cell infiltration and the lipid droplets, the tissue slice of liver and fat tissue and HE straining are made. Blood triglyceride(TG)、total cholesterol(CHOL)、high density lipoprotein(HDL-C) and low density lipoprotein(LDL-C) are tested. The statistical pattern we used is X±s, and we take SPSS19.0to do the variance analysis. p<0.05means statistical significance.Results:1、The body weight and the blood glucose of group high fat rise obviously compared with NC(p<0.05). The OGTT test indicates that group high fat mice are ill with type2diabetes. The blood glucose of group CD mice decrease obviously, while the blood glucose of group EKS is normal and even lower than group CD which means that the group EKS mice do not get the the type2diabetes. The blood glucose of group LKS has no difference with group HF mice(p>0.05).2、The mRNA levels of IL-6、IL-1β、TNF-a are also obviously higher in liver、 fat、muscle tissues in group high fat than in group NC(p<0.05). After treated with "tonifying kidney and strengthening spleen" and" clear heat and dry dampness",the mRNA levels of IL-6、 IL-1β、TNF-α liver、fat、muscle tissues also decrease largely in group EKS、group LKS and group CD mice than group HF mice. Compared with group EKS mice, the level of inflammatory factors decrease more in group CD mice.3、We find bigger adipose cell、more Inflammatory cell infiltration in tissue slice of fat tissue of group high fat than in group NC, and there are also much more lipid droplets in liver tissue slice of group high fat than in group NC. When treated with "tonifying kidney and strengthening spleen" and" clear heat and dry dampness", the lipid droplets become much fewer in liver tissue flice and the adipose cell become smaller in group EKS and group CD mice than group HF mice. The lipidosis remit more or less in group LKS mice liver, and the adipose cells also become a little smaller than group HF mice. Conclusion:1、It is feasible to construct type2diabetes model of mice by feeding mice with high fat food.2、If treated with "tonifying kidney and strengthening spleen "early to strengthen the foundation of power, there is no pathogenic factor formed and people do not get type2diabetes. Meanwhile the "tonifying kidney and strengthening spleen" can also protect our organs, so before people get types it is important that treating before getting ill.3、When people get type2diabetes characterized by fat, most people are Hot and humid remedying, and syhdrime strong for whom clearing heat and drying dampness may be better. It shows good effect on blood glucose after Type2diabetes mice are treated with "clear heat and dry dampness". The mRNA level of inflammatory factors decrease which then improve insulin resistent(IR). Through the mRNA level of inflammatory factors also decrease and the IR get improvement, the which then improve insulin resistent(IR). Through the "clear heat and dry dampness" show better result. All these results show the importance of treatment based on syndrome differentiation. We can take the right therapies only after we distinguish the symptoms and root cause which are the primary.
