Purpose:Investigate the relationship between neural apoptosis and the expression ofNF-κB/P65and cleaved-PARP after hypertensive cerebral hemorrhage.Methods:Collect varied part of tissue samples around the hematoma,stored in formalin andparaffin section; the surface part of the hematoma was group A, and semi-dark zonewas group B, the cortex was group C, both group A and B were experiment group,group C was the control; and we further subgrouped in the three groups accordingthe bleeding time(>7h,≤7h), bleeding volume (>60ml,<60ml).Test the expressionlevel of NF-κB/P65and cleaved-PARP in different hematoma areas after earlyhypertensive cerebral hemorrhage, detect the apoptosis rate of neural cell at vary partafter brain bleeding by TUNEL, analysis and explain the correlation between neuralcell apoptosis and the expression of NF-κB/P65and cleaved-PARP at thehypertensive cerebral hemorrhage through SPSS13.0.Results:1.Apoptosis cells were strong positive at the surface of hematoma detected byTUNEL, the further distance from the hematoma, the lesser the neural cells apoptosis.Statistic analysis: Group A:36.30%±2.83%,Group B:22.55%±3.49%,Group C:17.70%±2.62%,p<0.01.2.The expression of NF-κB/P65and cleaved-PARP significantly increased and strongpositive at the surface of the hematoma, the expression was obviously lesser inischemic hemi-dark band compared with the surface at hematoma, and the expression at the area of pallium was the lowest.3.The expression of NF-κB/P65and cleaved-PARP increased and positive rate raisedwhen the volume more than60ml.(NF-κB/P65: Z=-4.678,P<0.01, cleaved-PARP:Z=-6.036, P<0.01,TUNEL po:Z=-2.323,P=0.020<0.05).However when bleeding over7hours, the expression of NF-κB/P65,cleaved-PARP and positive rate of TUNELwere higher than patients who less than7hours(NF-κB/P65: Z=-4.327,P<0.01,cleaved-PARP:Z=-4.416, P<0.01,TUNEL po:Z=-2.252, P=0.031<0.05.)Conclusion:1,After HICH cell apoptosis around hematoma, the nearer from the hematoma,the more cells apoptosis.2,After HICH NF-κB/P65in cerebral tissue might promote neural apoptosisthrough caspase3represented by cleaved-PARP.3,As bleeding time and volume increased, the expression of NF-κB/P65andcleaved-PARP ascended, and worse the apoptosis.
