Experimental Study Of Apoptosis Expression In Rat's Brain Tissue With Hypertensive Cerebral Hemorrhage And Early Treatment With Hyperbaric Oxygen

Objective To investigate the apoptosis expression change with time in rat' s bilateral cerebral cortex and corpus striatum after hypertensive cerebral hemorrhage (HCH) and the protective effect of early treatment with hyperbaric oxygen (ETHO) to apoptosis damage in HCH.Methods 156 adult female Wistar rats were randomly divided into normal control group, false operation group, HCH natural recovery group and HCH-ETHO group. In model rats, the anterior branches of bilateral renal arteries were cogulated partly, then the animals were raised for 6 weeks with high salt, fat and cholesterol food, then intracerebral hemorrhage was induced by injection of bacterial collagenase mixed with hyparin into the right caudate nucleus. In false operation group, normal saline instead of bacterial collagenase and hyparin was injected into normal rats' caudate nucleus , and in normal control group the animals were nomal rats. Hyperbaric oxygen treatment was given at 4h after HCH in HCH-ETHO group. By TUNEL method, the apoptosis cells were measured in rats' brain tissues.Results In both normal control group and false operation group, only few of apoptosis cells appeared, showing no significant difference(t=0. 54~ 0.61, P>0.05). In natural recovery group, the apoptosis cells , at 4h~ 14d , expressed markedly more than the ones of both normal control group and false operation group, showing significant difference(t=9. 18~42. 56, P< 0. 05) ;and at 4h~7d , the apoptosis cells at hemorrhagic hemisphere expressed markedly more than non-hemorrhagic hemisphere, showing significant difference(t=2. 71~18. 39, P<0. 05) ; and the apoptosis peak of hemorrhagic hemisphere appeared at 18h ~ 24h, being earlier by 6 hours than non-hemorrhagic hemisphere .which appeared at 24h~36h, then both peaks decreased little by little to level of 6h at 14d;and at hemorrhagic hemisphere, the apoptosis cells in corpus striatum expressed more than the ones incerebral cortex, showing significant difference(t=2. 53~13. 84, P<0. 05) ; and at non-hemorrhagic hemisphere, the difference of apoptosis cells between corpus striatum and cerebral cortex was not significant (t=0.63 ?1.72,P >0. 05). In HCH-ETHO group, the apoptosis cells in both cerebral cortex and corpus striatum, at 12h~3d, expressed greatly less than natural recovery group .showing significant difference (t=2. 07~15. 34, P<0. 05), and ETHO had no obvious affect to the apoptosis peak.Conclusion In rat HCH, the apoptosis damage in hemorrhagic hemisphere is more severe than non-hemorrhagic hemisphere, and the apoptosis peak of bilateral hemispheres appears at different time. All anti-apoptosis clinical treatments to HCH should be given before 18h after HCH onset. ETHO has evident effect to decrease apoptosis expression in HCH, so it is a prospective medical strategy and worthy of further investigation.
