Objective To clarify the molecular mechanism of Chaipu decoction regulating theexpressions of T-lymphocyte maturation associated protein T(MAL) on airway epithelialcells,to elucidate whether Chaipu decoction up-regulating the expressions of MAL can reducesecretion of TGF-β1.Method32healthy male SD rats,weight (250±10) g,were assigned randomly intohigh dose (3.00g/kg)of Chaipu decoction group, medium (1.50g/kg), low (0.75g/kg) dosegroup, and saline group,with8rats in each group. Rats from homologous groups were givendifferent doses of Chaipu decoction or saline solution twice per day for7days by intragastricadministration.After treatment, blood was collected from rat heart, and serum was prepared.32healthy male SD rats,weight (250±10) g,was taken to establish a mice asthmatic model andasthmatic serum was prepared, the lung tissue was obtained and stained by HE. Asthmaticmodel was established through the OVA together with aluminum hydroxide sensitization andatomization. Rat tracheal epithelial cells were devided into Group A(normal serumgroup),Group B (Chaipu decoction high dose+asthma serum group),Group C (mediumChaipu dose+asthma serum group),Group D (medium Chaipu dose+asthma serumgroup),Group E (saline serum+asthma serum group),Group F (asthma Serumgroup). Theexpression of MAL,TLR-4,NF-κB,TGF-β1in the different groups was measured by usingRT-PCR and immunohistochemical method.ResultThe pathologic changes of rat lung tissue.In control group,bronchial mucous membrane presented continuous structure,noinflammatory cell infiltration,pulmonary alveoli intact wall.Bronchiolar dilatation and wallthickening,the bronchial lumen narrowing,Most alveolar damage, fusion, alveolar spaceenlarged, alveolar interval fracture can be seen in the asthma group. Expression levels of MAL,TLR4,NF-κB,TGF-β1in the cell.The MAL mRNA and protein expression levels in Group A were higher than othergroups(P<0.05), while in Group E,F declines significantly with asthmatic serum intervention.The expression levels of MAL mRNA and protein in Group B,C, D(with Chaipu Decoctionserum intervention) were significantly higher than those in Group E, F (P<0.05), significantdifferences were found between MAL levels in Group B,C,D and Group E,F(P<0.05).Conversely, the expression levels of the TLR-4,NF-κB,TGF-β1mRNA and protein inGroupA are in the minimum amount, compared with other groups, there are very significantdifferences(P<0.05). The expression levels of TLR-4,NF-κB,TGF-β1mRNA and protein inGroup B,C, D(with Chaipu Decoction serum intervention) were significantly lower than thosein Group E, F with asthmatic serum intervention. The TLR-4,NF-κB mRNA and protein werehighly expressed in Group E,F were significantly higher than those in Group B,C,D(P<0.05),and the expression of TGF-β1mRNA and protein in Group E,F were significantly higher thanthose in Group B,C(P<0.05).Conclusion:1. Asthmatic secrum could decrease the expression of MAL and increase the expressionof TLR-4,NF-κB,TGF-β1in airway epithelial cells.2. Chaipu decoction can regulate the expressions of MAL and TGF-β1, the mechanismmay be associated with TLR-4/NF-κB.
