Objective: Observed in the rat model of asthma TLR4, NF-?B, MAL and TGF – ? 1 expression level, analyze its and the influence on airway remodeling, the relationship between explicit Chaipu decoction can in the rat model of asthma by TLR4/ the nf-kappa B pathway MAL expression. Methods: 32 healthy male SD rats, the quality of body?20? 250 + g, randomly divided into 4 groups, the normal group, PDTC group, Chaipu decoction group, asthma group each group of eight, with egg protein replication asthma model, abdominal aorta after excitation bleed to death, adopt the method of HE staining to observe each group pathological changes, and using image software testing lung airway morphology parameters; Using reverse transcription polymerase chain reaction?RT-PCR? and immunohistochemical detection of airway TLR4, NF-?B, MAL and TGF – ?1 expression level. Results: 1. The rat lung tissue HE staining and airway parameters:Under a microscope and HE staining observation, airway wall full visible to normal group, no obvious inflammatory cells in the surrounding invasion and goblet cell hyperplasia; Asthma groups, Chaipu decoction group, PDTC group compared with normal group of airway wall thickening obviously, bureaucratic narrow, alveolar fusion is more, significant difference was statistically significant?P < 0.05?;Chaipu decoction group airway wall thickness in the asthma group and PDTC group were lower than the asthma group, the difference between comparison was statistically significant?P < 0.05?, the comparison between Chaipu decoction group with PDTC group, there was no statistically significant difference?P > 0.05?.2.RT-PCR Method to measure the expression of TLR4, NF-?B, MAL and TGF – ?1:RT-PCR bands show in the normal group TLR4, NF-?B, TGF – ?1 mRNA express the lowest in the normal group, there was a significant difference compared with the other groups?P < 0.05?; Asthma group of TLR4, NF-?B, TGF – ?1 mRNA expression and the rest of the group, there was a significant difference?P < 0.05?; Chaipu decoction group with PDTC group TLR4, NF-?B, TGF-B compared to beta mRNA expression, no obvious difference, no statistical significance?P > 0.05?. The highest MAL mRNA expression in normal group, and the rest of the group than there was a significant difference?P < 0.05?; MAL mRNA expression in asthma group than other group significantly decreased, there was a significant difference?P < 0.05?; MAL mRNA in between Chaipu decoction group with PDTC group no significant difference?P > 0.05?.3. Immunohistochemical detection of each group in rat lung tissue TLR4, NF-?B, TGF – ?1 protein expression:Normal group of TLR4, NF-?B, TGF – ?1 protein expression of the lowest in the normal group, there was a significant difference compared with the other groups?P < 0.05?; Asthma group of TLR4, NF-?B, TGF – ?1 protein expression and the rest of the group, there was a significant difference?P < 0.05?;Chaipu decoction group with PDTC group TLR4, NF-?, TGF-B compared to beta protein expression, there is no obvious difference, no statistical significance?P > 0.05?. The highest MAL proteins expressed in the normal group, and the rest of the group than there was a significant difference?P < 0.05?; MAL protein expression in the asthma group than other group significantly decreased, there was a significant difference?P < 0.05?; MAL proteins in between Chaipu decoction group with PDTC group no significant difference?P > 0.05?.Conclusion: 1. TLR4, NF-?B pathways involved in the process of airway remodeling of asthma. 2. Chaipu decoction can increase MAL expression in the airway, the mechanism may be mediated TLR4/NF-?B signaling pathway activation.
