| Purpose:By cultivating primary knee osteoarthritis synoviocyte, to observe the effect of serum of Duhuojisheng Decoction plus astragalus on TNF-α and IL-6, to explore the reaction mechanism of knee osteoarthritis and the curative effect of Duhuojisheng Decoction plus astragalus on knee osteoarthritis, so that scientific grounds for the curative effect of Duhuojisheng Decoction plus astragalus on knee osteoarthritis is offered.Methids:SPF-grade male SD rats were divided into3groups and were given by gavage Duhuojisheng Decoction plus astragalus, Meloxicam, and physiological saline respectively. After3days blood samples were obtained from abdominal aorta to make serum. The cultivated P2-generation knee osteoarthritis synoviocyte were under the intervention of the serum of Duhuojisheng decoction plus astragalus (experimental group), Meloxicam serum (control group) and physiological saline serum (blank group) respectively for24h to see their effect on TNF-aand IL-6in knee osteoarthritis synoviocyte via qRT-PCR and ELISA.Result:By qRT-PCR testing found that the Chinese group the expression of IL-6and TNF-amRNA was0.53±0.082,0.68±0.054, nearly50%of the control group, the WM group were0.27±0.117,0.37±0.093, the equivalent of a blank35%of the group (P<0.05). By ELISA test,-6IL expression of the Chinese group and TNF-a protein were21.06±1.427ng/ml,37.62±1.970ng/ml, as compared with the control group decreased by approximately30%, Western medicine group were12.86±3.619ng/ml,24.77±3.024ng/ml, reduced50%(P <0.05), but the medicine group and the Chinese group decreased significantly compared, there was no statistically significant difference (P>0.05).Conclusion:Duhuojisheng decoction plus astragalus inhibits the expression of TNF-αand IL-6in P2-generation knee osteoarthritis synoviocyte. By controlling inflammation reaction, Duhuojisheng decoction plus astragalus reduces the knee-joint ache and improves knee-joint function, and knee osteoarthritis is in this way treated. |
