Objective This experiment was to study the relationship between methylation level of lipid metabolism related genes and the cerebral infarction togerther with its related risk factors.Methods By adopting the case-control study method,we randomly recruited fifteen cerebral infarction patients and fifteen healthy controls from the han population of hunan.DNA methylation in the promotor region of LPL gene were measured by bisulfite sequencing PCR(BSP).Results1. Compared to the control group, the diastolic blood pressure, low density lipoprotein cholesterol levels of the case group were higher than the control group, HDL cholesterol levels of the case group were lower than the control group.The difference was statistically significant (P<0.05).2. Compared the average level of methylation(CpG%) between the two groups of each CpG sites, CpG9to CpG16as well as CpG20to CpG21of the case group were obviously higher than that of the control group, the difference was statistically significant (P<0.05). The average methylation level of total thirty CpG sites(CpGZ%) in LPL gene promoter region was significantly higher than the control group, the difference was statistically significant (P<0.05).3. The MRI examination results of15patients and the carotid colorized ultrasound results of25pepole were recorded. Compared the lacunar cerebral infarction group and the group of atherothrombotic cerebral infarction, the overall average LPL gene promoter methylation levels(CpGZ%) had no significant difference (P>0.05).Compared the carotid atherosclerosis group and the carotid atherosclerosis not found group, the overall average LPL gene promoter methylation levels(CpGZ%) had no significant difference (P>0.05).4. The high-density lipoprotein cholesterol levels in all studies and the overall average LPL gene promoter methylation levels(CpGZ%) had significant negative linear correlation (P=0.001). At the CpG9to CpG16sites the linear correlation was significant, the difference was statistically significant (P<0.05). The LDL cholesterol levels in all studies and the overall average LPL gene promoter methylation level(CpGZ%) had significant positive linear correlation (P=0.004).At the CpG10to CpG12, CpG15、CpG20sites the linear correlation was significant, the difference was statistically significant (P<0.05).5. There was significant positive linear correlation between age and the overall average LPL gene promoter methylation level(CpGZ%)(P=0.024),the difference was statistically significant at the CpG9to CpG16sites(P<0.05).6. The overall average LPL gene promoter methylation level(CpGZ%) were compared between the cases and the controls respectively in male group and female group. LPL gene promoter overall mean methylation level(CpGZ%) of the male cases and the male controls had significant difference (P=0.001),as well as the female cases and the male controls (P=0.024).Conclusion1.The LPL gene promoter region hypermethylation was associated with atherothrombotic cerebral infarction, as well as the low HDL-C concentrations and high LDL-C concentrations.2.The LPL gene promoter region methylation levels might have no influence on the infarction area and carotid atherosclerosis.3.The LPL gene promoter region methylation levels had increased with age. The LPL gene promoter region methylation levels had no gender differences.
