BackgroundHuman apolipoprotein E(ApoE)gene is a key gene for liver to remove residual lipoprotein,a kind of important gene being resistant to atherosclerosis ?Apolipoprotein E(ApoE)gene and its protein(ApoE)play a key role in lipid metabolism,including the redistribution of lipoprotein and cholesterol?ApoE is divided into three common subtypes: E2?E3 and E4,respectively coded by epsilon 2?epsilon 3 and epsilon 4 three alleles?The survey of large crowds suggests epsilon 4 alleles can significantly increase healthy persons total cholesterol level,making carriers be susceptible to atherosclerosis;on the contrary,epsilon 2 allele can reduce carriers' cholesterol level,and its lowering cholesterol effect is 2-3 times of epsilon 4 elevateing cholesterol effect?In addition,the epsilon 2,epsilon 3 and epsilon 4 alleles of ApoE are associated with familial and sporadic Alzheimer's Disease(AD)?With an aging society,there is an upward trend in the incidence of cerebral infarction;especially,atherosclerotic cerebral infarction is the most common,accounting for about 60% of the total cerebral infarction,is the focus of this study?Cerebral infarction is a kind of polygenic inheritance disease,the result of inherited and environmental factors interacting and epigenetics provide a new idea for research about it?Epigenetics refers to in the premise of same gene sequence,gene expression changed heritably,including DNA methylation?histone modification?chromatin remodeling?non-coding RNA regulation and so on,and the research of DNA methylation goes deeper into?Based on the above,this study aims to explore the correlation of the ApoE gene promoter region DNA methylation and the atherosclerotic cerebral infarction? PurposeExplore the correlation of the ApoE gene promoter region DNA methylation and the atherosclerotic cerebral infarction? MethodWith case-control study,we randomly extracte henan han nationality population,a total of 52 cases,26 atherosclerotic cerebral infarction patients and 26 healthy controls,detecting their ApoE gene promoter region DNA methylation status? Result1.Hypertension and carotid atherosclerosis plaque formation have significant differences(P<0.05)between the two groups,the case group obviously higher than the control group?Comparing with the control group,high density lipoprotein cholesterol(HDL-C),folic acid in the case group are significantly reducer,with statistical significance(P<0.05)?Homocysteine level of the case group is higher than the control group,with significant difference(P<0.05)?2.Methylation of CpG14?CpG16 have significant differences(p<0.05)between the case group and the control group?3.After adjustment for age ? gender and other covariate,CpG16 ? carotid atherosclerotic plaques and folic acid are significant between the case group and the control group,the OR values are respective 16.146(95%CI: 1.154~225.832),27.194(95%CI: 2.266~326.362),0.586(95%CI: 0.355~0.968),suggesting about the risk of atherosclerotic cerebral infarction,people with CpG16 methylation are 16.146 times of people without CpG16 methylation,and the crowd of carotid atherosclerotic plaques are 27.194 times of people without carotid atherosclerotic plaques?Within a certain range,the content of folic acid is higher,the risk of atherosclerotic cerebral infarction is lower? ConclusionApoE gene promoter region DNA methylation,carotid atherosclerotic plaque can increase the risk of atherosclerotic cerebral infarction,folic acid is the protective factor of preventing from atherosclerotic cerebral infarction?...
